Study On Preparation Technology And Quality Standard Of Xiangyu Cataplasma

Pedo-diarrhea is clinical frequently encountered disease. Because of the difficulty of children Medication, recent reseaches are focus on external preparation. The merits of cataplasma include repeated use, contain more medicine etc, which make them more perspective clinically. The prescription includes six kinds of herbal medicine, such as Radix Codonopsis, Rhizoma Atractylodis, Radix Linderae, Fructus Evodiae, Herba Pogostemonis and Galla Chinensis, which have the effect of invigorate the spleen and deprive the evil wetness to check diarrhea.Arguing the rationality of the cataplasmata containing Traditional Chinese Medicine, this work had developed a new preparation of Chinese Medicine which is safety and effective on pedo-diarrhea, based on the study about the Xiangyu cataplasma.By means of orthogonal design, the extraction technology of volatile is optimized with the yield of volatile oil as an index. According to the content of the evodiamine as an index, the extraction of Fructus Evodiae is optimized with alcohol deposition, and the extraction of other medicinal substances is developed with water deposition by the content of galic acid which is regarded as another index. Meanwhile, depurate technology is determined by HPLC. The optimum proportion of matrix for the preparation technology process of cataplasma is selected by homogeneous design. Molding of cataplasma is identified. As a result, the best methods to extract the six kinds of medicine as follows, Radix Codonopsis, etc. distilled with water for volatile in four hours, then the residue are extracted by 10 times volume water for two times. After purified by alcohol, the combining part is concentrated into extractum. The best extracting approach for Fructus Evodiae is eight times volume of 90 percent alcohol for three times.The optimum proportion of matrix for the preparation technology process of Cataplasm is selected by uniform design and SPSS regression analysis. A check-up for adhibition , peeling strength, nonflowing, content of cream is founded. The best matrix's prescription is gelatin : CMC-Na : PANA : kaolin : aluminum trichloride : citric acid : PVP K-30 : PEG400 : trimethylene glycol: tween-80 is 0.25 : 0.1 : 0.25 : 1.5 : 0.4 : 0.6 : 0.8 :1: 0.5 : 0.25.In the research on the quality standard, Radix Codonopsis, Rhizoma Atractylodis, Radix Linderae and Herba Pogostemonis are qualitatively tested by TLC. By using HPLC, we measure the contents of the galic acid and evodiamine in the drug. Meanwhile, we discuss on the methods of extraction and reseach the technology. On one hand, the study results of the quality standard show that during the TLC process, the four kinds of herb medicine have good resolution and specificity. On the other hand, the contents of galic acid and extractum are accurate and have good reproducibility. The quality standard can be used to control the quality of Xiangyu cataplasma The preparation technology of cataplasm is feasible, and its quality is controllable, so it is an stable transdermal-drug delivery system.
