Extraction Of Jujubosides By Enzymatic Method

Semen Ziziphi Spinosae is the dried ripe seed of Ziziphus jujuba Mill.var.spenosa(Bunge) Hu ex H.f.Chou and is defined as a good medicine in Compendium of Materia Medica.It is sour in taste and neutral in nature and is distributed to the liver,heart and gallbladder channels.It can replenish the liver,calm the nervous system,arrest excessive perspiration and promote the production of saliva and has been traditionally used to treat insomnia,dreaminess,excessive perspiration and thirsty.Pharmacological studies have shown that Saponins extracted from Semen Ziziphi Spinosae has favorable efficacy in insomnia and neurasthenia induced by various causations,with low toxicity and rare side effects.So it is a natural sedative and hypnotic traditional Chinese medicine,with a better security than that of Western medicine.In order to increase the content of jujubosides in the extract of Semen Ziziphi Spinosae,the extraction of jujubosides by enzymatic method compared with traditional water extraction, ethanol extraction and semi-bionic extraction method were performed.The optimal technique condition of jujubosides extraction by enzymatic method was investigated and purificatory technique by ultrafiltration and macroporous adsorption resin was studied.With jujuboside A as the comparison,the extract was validated by TLC.The saponins extracted by enzymatic method met the criterion of standard product.The sedative and hypnotic effect of the extract was simultaneously observed.The results are as follows:1.The extraction of jujubosides by enzymatic method is the best extraction method.The optimal technique condition of jujubosides extraction by enzymatic method was investigated.The optimum conditions included 40℃for enzymolysis,4.5 in pH value of the initial enzymolysis liquid,cellulose enzyme at 30U/g,compound pectic enzyme at 30U/g,and 5h for enzymolysis.2.With jujuboside A as the comparison,the extract was validated by TLC.The saponins extracted by enzymatic method met the criterion of standard product.3.The extract of jujubosides was refined by ultrafiltration and the macroporous adsorption resin,and the purity of the extract of jujubosides by enzymatic method was up to 48.96%.4.The pharmacological study showed that jujubosides extracted by enzymatic method had preferable regulatory effect on the dormancy of mice.They can increase the sleeping rate of mice treated with low dose Sodium Pentobarbital,remarkably shorten the sleeping latency,and prolong the sleeping time. 5.An UV spectrophotometry method was established to determine the content of jujubosides.The spectrums of jujuboside extract with different techniques were studied by HPLC-ELSD,which laid a foundation for establishing the quality standard of jujubosides extracted from Semen Ziziphi Spinosae.Conclusion The extraction rate of jujubosides by enzymatic method was higher than that of by water extraction and ethanol extraction.The extraction which applies enzyme biological technology to get jujubosides from Semen Ziziphi Spinosae is an effective method.
