Hepatitis B is one of the major viral infectious diseases, which are harmful to human health. Vaccination can effectively prevent transmission of the virus. In China the infection rate of Hepatitis B is 60% and the HBV carrier rate is 10% (equals to 0.12 billion). There are 0.3 million patients die of HBV related diseases each year. Because of the high infectivity, large infection area, and high pathogenicity, HBV causes huge damage in physical and mental healthe as well as economy.Hepatitis B vaccine plays an important role in the prevention of hepatitis B The most important step during the large-scale production of and yields more social and economic benefit. The most important step during the large-scale production of Hepatitis B vaccine is purification which directly affects the purity and recovery of the product. The objective of the experiment is to optimize the density gradient centrifugation method, and discuss slightly about crude extraction method. In order to establish a low-cost, high recovery purification process, the optimization includes the bacteria selection, the choice of appropriate ultra-centrifugation method, changing the gradient ratio of the The most important step during the large-scale production of solution, increasing the purity and yield of products etc.
