| The pellet of shengxue is composed of Radix Astragali, Radix Angelicae Sinensis, Radix Et Rhizoma Ginseng, Radix Rehmanniae, Radix Paeoniae Alba,Rhizoma Chuanxiong,Rhizoma Curculiginis, Herba Epimedii, Poria, etc. It has many pharmacologic actions, such as nourishing vigor and recuperating blood, nourishing Y in and kidney, flowing blood and caring spleen. It's used in the treatment of blood-debilitated, and at present it is extensively used in the treatmeat of regeneratively obstructive anaemia. The original prescription derived from the book Yi Zong Jin Jian". In our search, Rhizoma Curculiginis, Herba Epimedii, Poria were added in the pellet. This paper was focused on extraction, preparation, identification in qualitation, fixed quantity analysis and inspection in quality.In order to extract much more active substance, the extraction methord of volatile oil in Radix Angelicae Sinensis and Rhizoma Chuanxiong followed by water was used. In volatile oilextraction, orthogonal experimental design was applied to optimize the process parameters which affected result of extraction seriousely such as time of soak, time of distillation and volume of water. Based on the volume of volatile oil, the optimal extraction was as follows: distill medicinal herbs with ten times of water for 4h. Three main factors such as time of extraction, volume of water and times of extraction were considered. The research on the criterious of the contents of Astragaloside IV showed that the best extraction technology was ten times of medical residue, and extract three times and 1h per time. Decompression condense and decompression dry technology were used to make sure active substance was not destroyed. Temperature, pressure, relative density and speed which material comes into tower were optimized in the process of condense and dry.The variety and quantity of adjuvant were screened on the basis of hygroscopicity in preparation technology. The result showed that the best adjuvant was dextrin and the desirable quantitu was 0.3 times of extractum. We prepared shengxue pellets according to preparation technology and inspected a series of character. The result were as follows: bulk density was 0.6316g/ml, angle of repose was 33.7°.The results indicater that fluidity of pellets was good and humidity should be controlled below 55% in case of excessive water in pellets.During quality research, Radix Astragali, Radix Angelicae Sinensis, Radix Et Rhizoma Ginseng, Radix Rehmanniae, Radix Paeoniae Alba,Rhizoma Chuanxiong, Rhizoma Curculiginis, Herba Epimedii, Poria, in this prescription were identified by TLC. The methods and chromatogram were provided. Negative was not disturb medicial and spot of active substance was very clear. According to standard on version 2005 of Chinese pharmacopeia, paritcle size, content uniformity, moisture, and capacity of dissolving and microbial limit were inspected. The results showed that three batch of samples were up to standard. A HPLC method for determination of astragaloside IV in the pellerts wasestablished. A good linear relationship between Injected Sample Volume and Natural Logarithan of peak area of astragaloside IV was found within the range of 2.024~10.12ug(r=0,9998), the average recovery was 99.42% and RSD=1.59%. This method was found to be sensitive and accurate for the measurement, and can be used for the quality cntroll of shengxue pellets. The results of three batch of samples indicated limit of astragaloside IV shouldn't be lower than 7.0mg per bag. |
