Studies On Preparation Process And Quality Standards Of Kudingcha Tablets

Ilex kudingcha C.J Tseng is used as traditional planta medica beverage in China.It is also used in popular medicines and commercial herbal preparations as a stimulant to central nervous system,a diuretic,a treatment for sore throats,an aid to losing weight and relief of hypertension.The research of its pharmacology and health-pretection fuction is based on traditional water solutions.The toxicology research disclose:Kudingcha water extrations has no toxin properties;it is safe to take and it is appropriate to use as health-protection food.Considering its bitterness,this experiment makes Kudingcha water extractions into tablets,which is convenient to carry about.The author has made a study on its preparation and quality standard.ⅠStudy on the preparation1.In accordance with demand of health-pretection food,with water as a solvent,using astragalin and the transferring rate of total saponin as evaluating indexes,we selected whether soaking or not to extract by contrast design,and we find there is no statistical sense on the bases of experimental result analysis.Therefore we select the method with no soaking.Using solvent volume,extracting time and extracting times as examing factors,using astragalin,the transferring rate of total saponin and Kudingoside D as evaluating index,we selected the opimum water extracting condition by orthogonal design,which is:extracting three times, adding 15 times solvent volume,extracting one hour per-time.2.After condensing the water extracting solutions,using the content changes of total saponin and Kudingoside D as indexes,we study on the process of dryness by contrast design.The optimum vaccum dryness temperature is 80℃.3.Contrast experiment is carried out on the adjuvant and the manufacturing process of tablets.According to experiments results,we can make up the type and the dosage of adjuvant,also the manufacturing process is ascertained for Kudingcha tablets.4.In order to increase the steadiness of the tablets,film coating experiment is carried out successfully. ⅡStudy on quality standard1.With Kudingoside E and Kudinogside D as standard contrast,Kudingcha was identified by TLC.2.An UV method for the determination of total saponin and a HPLC method for the determination of Kudingoside D were established.Assay methods were validated in the research of content.Average recovery rate was 100.48%while RSD was 2.55%in the UV method and this method was proved to be simple,accurate and reliable,which may be used for the quantitative determination of total saponin in its preparations.
