Fructus Gardeniae(TCM) is dry and ripe fruits of Fructus Gardeniae which belongs to rubiaceae plant.Fructus Gardeniae contains the following chemical ingredients:Lridoid Glycosides(Geniposide),Flavonoid(Gardenin),organic acetate (chlorogenic acid,croceic acid) etc..Fructus Gardeniae can protect liver,brain tissue and pancreas cells,also can accelerate bile excretion,adjust functions of stomach, increase PVI,repair orthopaedics soft tissue injury,heart hypotensive,antimicrobial effects and inflammatory which shows extensive medicinal value.In this experiment,10 batches of Fructus Gardenia samples were collected,the best extraction technology was determined.Carry out the method for high performance liquid chromatography,taking research on the fingerprint spectrum and its relativity of the medicinal materials and extracts,this experiment supplied a significative exploration for evaluating the quality of the medicinal materials and formulations scientifically.Fructus Gardenia contains hydrosoluble components,such as Iridoid Glycoside, Gardenoside and genipin gentiobioside,so water,50%methanol and methanol are used as extraction solvent in the test preparation.After comparing test results,all the spectrums are similar as others.Water solution is very difficult to filter,and 50% methanol has low toxicity,so 50%methanol is used as extraction solvent in the test preparation.Results of HPLC indicate that the repeatability of this extraction method is good.Choose geniposide which has a clear structure as reference,use a column packed with Kromasil C18,detection wavelength is 254nm,as a result,HPLC fingerprint spectrum is gotten which can give a true reflection of the Fructus Gardenia quality.The spectrum gotten in this experiment shows that the chemical fingerprint information is full,the resolution of the peaks is good with obvious characteristics. The precision,stability and repeatability of this method meet the requirements of the research on fingerprint spectrum,and this method can be used as a scientific evidence of evaluating and controling the Fructus Gardenia quality.According to the property of Fructus Gardeniae,its extraction technology design water extraction,ethanol precipitation,1-butanol extraction.The major factors influencing water extraction technology are water quantity,herbs soaking time, decoction frequency and decoction time.The best fructus gardeniae extraction technology is confirmed to 10 times quantity water than fructus gardeniae herbs, decoction 2 times,each time 2 hours by orthogonal test in four factors and three levels L9(34).After pilot production,the technology is proved to be reasonable,stable and practicable.The major factors influencing ethanol precipitation technology are extract concentration,extract temperature,ethanol precipitation concentration and laying aside time.The condition for first ethanol precipitation are decoction liquid concentration(per ml containing 1.0g of dried medicinal herbs),extracts cooling to room temperature(20~25℃),precipitate with ethanol,make it become 75%ethanol solution,stand for 12h by orthogonal test in four factors and three levels.After pilot production,the technology is proved to be practicable.The condition for second ethanol precipitation are first ethanol precipitation concentration(per ml containing 2.0g of the dried medicinal herbs),extracts cooling to room temperature(20~25℃), precipitate with ethanol,make it become 85%ethanol solution,stand for 12h.After pilot production,the technology is proved to be practicable.The major factors influencing 1-butanol extraction technology are medicine liquid concentration (containing dried medicinal herbs quantity),solvent quantity and extraction frequency. The best conditions for 1-butanol extract technology are medicine liquid concentration(equals to per ml containing 2.0g dried medicinal herbs),extract 4 times with 1.5 times of 1-butanol quantity.After pilot production,the technology is proved to be practicable.Ten batches of extract prepared by extraction technology according to this thesis, after measured by fingerprint chromatography,which shows technological conditions are steady and practicable,the repeatability is good,the relevance between fingerprint chromatography of extract and that of medicinal materials is good,main feature peaks of medicinal materials are shown in the extract chromatography.Fingerprint chromatography established in this thesis and extraction technology are all applied well in commercial manufacturing,especially the quality of medicinal material can be controlled fingerprint chromatography and production process can be followed so that the preparation's quality will be improved a lot.
