Marigold (Tagetes erecta L.) originating in Mexico, also known as Calendula officinalis, Hibiscus smell is rich in lutein, flavonoids.It is widely used in foods, cosmetics, pharmaceutical, tobacco, poultry feed and other fields.This paper discussed the ultrasonic extracting lutein esters through the single-factor tests and orthogonal test, four parameters of extraction time, extraction temperature, ratios of material to liquid, ultrasonic power affecting the yield of lutein esters. The experimental results show that ratio of material to liquid affected the yield of lutein esters significantly, optimal conditions of ultrasonic extracting lutein ester are as follows:extraction solvent, chloroform:tetrahydrofuran (V:V)= 1:1, ultrasonic power is 160W, liquid ratio of 1:7, temperature is 40℃, extraction time is 40min, solid to liquid ratio of 1:2, in the above-mentioned conditions, marigold flower extract lutein esters content was 17.17mg/g.Lutein is the main form of ester derivatives in the plant, it has to go through saponification treatment, and will be reverted to the monomer form of lutein. In this study, we speeded up the saponification time, lowered temperature saponification by ultrasonic 'cavitation' and mechanical action to. Using thin-layer chromatography, we inspected the effects of saponification time, saponification temperature, increasing the volume of alkali and the order of increasing alkali saponification to derive optimum saponification conditions:KOH concentration of 10% methanol, lye dosage of 30ml/ g, saponification time 3h, saponification temperature of 50℃.We studied the separation and purification of lutein by recrystallization the use of Lutein crude(g):tetrahydrofuran (ml):ethanol (ml):petroleum ether (ml):water (ml) =1:20:15:5:20 mixed solvents made the first recrystallization. Acetone, petroleum ether and water mixed solvent make the second recrystallization.Followed by a four crystalhigh purity lutein (78.40%)were made from the low purity of lutein,40.31% yield of compared to lutein ester. However, due to recrystallization to greater losses, long time, so choose salting purified lutein to 55% saturation were salting NaHSO4,lutein content reached 88.32%,1.63% yield of marigold lutein.This paper studied the lutein in the light, heat, acid and alkali under the conditions of the nature of existence changes, compared to its stability, and withβ-cyclodextrin to increase its water solubility, experimental results show that:light, high temperature (= 60℃), acid stability of large lutein; through the single-factor test, the feed ratio (lutein:β-CD), inclusion time, and the inclusion of temperature. The experimental results show that the inclusion time affected the yield of the inclusion significantly, optimal conditions of ultrasonic extracting Lutein-β-cyclodextrin are as follows:feed ratio (lutein:β-CD) was 1:2, the inclusion time 4h, inclusion temperature 35℃. Application of this technology by inclusion in the drug loading was 12.95%, the yield of indusion was 62.02%, the water solubility was 738.34mg/L.The orthogonal design was used in the research on the extraction technique of total flavonoids from Tagetes erecta with organic solvent assisted with microwave. The best extraction technique of total flavonoids from Tagetes erecta was explored. optimal conditions of ultrasonic extracting flavonoids are as follows:70% ethanol as extraction solvent, extraction temperature 70℃, microwave-assisted back to 60 min, microwave power 300 W, solid-liquid ratio of 1:8,2 times. By experiments, in these conditions, the marigold flower extract residue particles yield 15.86%, extract content was 44.01%,6.98% yield of total flavonoids.
