Aster ageratoides is also called ’Sanmaiye Malan’,which belongs to the Aster genus (Asteraceae). It has various medical functions such as moistening lung to lower qi, relieving cough and reducing sputum, clearing heat and removing toxicity et al, and usually it is used for treatment of innominate toxic swelling, hot cold in folklore. It is an effective TCM for remedy of cough and asthma in Jiangxi folelore, and so is also named ’Huanfeicao’. Researches on chemical constituents of A. ageratoides have been widely carried out so far, while researches on the extraction and purification technology of effective fraction and active componet ratio has not been reported. Therfore, in this paper we will report the optimum conditions for preparation technology of total flavones and total saponins from A. ageratoides and their propotion for anti-chronic bronchitis so as to lay foundations for industrial preparation of these active fractions and further developments of innovating medicine and health products. The major contents and conclusions in this paper were listed as below:1. The extraction and purification process of total flavones from Aster ageratoides has been studied in detail. The optimum extraction conditions were investigated by the contents of the total flavones, using orthogonal test and ethanol as extraction solvent. The optimum extraction conditions were as follows:using60%alcohol of ten times than a mounts of original material soaking2hours, extracting three times with hot reflux method at50℃. Static adsorption capacity and desorption rate were employed as examine items for the screening of optimum macro porous resin and optimum technology for the purification of total flavones with three commercial resin such as HPD-500、HPD-600、HP-20was investigated. The HPD-500type resin showed the best purification effect and was chosen as the material for the next step dynamic adsorption. The optimum purification conditions were as follows:the sample solution was prepared at the concentration of50mg/ml, subjected to HPD-500type resin column chromatography with a load ratio of16mg total flavones per gram of resin. After standing for1hour, the column was eluted with4BV water before being eluted with2BV70%alcohol.Under the optimal purification conditions, the purity of total flavones was76.7%, which enhanced3.83times.2. The preliminary research on extraction and preparation process of total saponins from A.ageratoides has also been carried out. The optimal extraction condition has been determined on the basis of orthogonal test. The optimum extraction conditions were as follows:using60%alcohol of eight times than a mounts of original material soaking1.5hours, extracting one time with hot reflux method at30℃. Static adsorption capacity and desorption rate were employed as examine items for the screening of optimum macro porous resin and optimum technology for the purification of total saponins with three commercial resin such as HPD-500、HPD-600、HP-20was investigated. The HPD-500type resin showed the best purification effect and was chosen for the next step dynamic adsorption. After standing for1hour, the column was eluted with4BV water before being eluted with4BV70%alcohol.Under the optimal purification conditions,and the purity of total sapanins can be raised2.67times to50.12%.3. The optimal composition for anti-chronic bronchitis from A. ageratoides has been determined. Different proportion of total saponins and total flavonoids has been setted with increase-decrease baseline geometric proportion design method, and the optimal composition was optimized through the expectorant cough experiment. For the same total dosage, the eliminating phlegm effects of different proportions of total saponins and total flavonoids of A. ageratoides were dose dependent with total saponins; the stopping cough effects of group1:1and group1:2were not only dependent with total flavonoids, but also dependent with synergetic compatibility relationship between total saponins and total flavonoids.
