In this paper, we studied the extraction technology for the total flavones from Viola yedoensis Makino and the adsorption and separation technology for chlorophyll from extracted solution. Methods: The concentration of total flavones and chlorophyll in Viola yedoensis Makino was determined with UV spectrophotometry. Orthogonal text and single-factor test was used to optimize the extraction of total flavones in Viola yedoensis Makino, which was measured against the concentration of total flavones extraction. The modified micro-powder TD-21 was used to absorb chlorophyll, with the recovery rate of total flavones and the remove rate of chlorophyll as indexes. We studied five factors which affect the result of experiment. According to the conclusion, the main factors which affect adsorption more are the adsorbent amount, pH and the concentration of chlorophyll. Results: After methodology inspection,Good linear relationship with the absorbability was achieved when the detection concentration of rutin was within a range of 0.012~0.120g/L, and the system serviceability was good. The optimum extraction process is as follows: 60% alcohol, 20 times volume of the weight of the dried material,refluxing 90min for 3 times,the concentration of total flavones was 3.20%. The optimum adsorption and separation process is as follows: 0.5g/50ml adsorbent amount, the concentration of chlorophyll was 6mg/L, pH=3, mixing speed is R2, adsorbing 30min, the recovery rate of total flavones was 90.82%, the remove rate of chlorophyll was was 90.82%. Conclusion: The method of UV spectrophotometry for determination of the total flavonoides is simple and accurate. This study explored a practical extraction method and separation methoand lay a good foundation for Viola yedoensis Makino research in the future.
