In this paper, extraction purification and antioxidation of polyphenols of from seed of Camellia oliefera polyphenols were studied. The conclusions of this research were as follows:Two methods to extract of polyphenols of from seed of camellia oliefera polyphenols were studied. The results of solvent extraction showed that the optimum conditions to extract polyphenols from seed of Camellia oliefera were:temperature 25℃; methanol:water(1:1); 60 min; ratio of materials and solution (1:15). the extraction rate of polyphenols is 18.34 mg/g. Furthermore, the optimum conditions for microwave assisted extraction (MAE) technique were obtained through Response Surface Methodology. The optimum conditions were:microwave radiation 800 W, extraction time 35 s, methanol concentration 60%, the ratio of material to liquid 1:16; the maximum extraction rate of polyphenols 19.02 mg/g. Under the same condition, comparing MAE technique and extraction with solvent extraction, the result showed that the effect of microwave extraction was better.The extraction yield of polyphenols of seed of camellia oliefera was infl-uenced by the variety, the habitat, storage time. in this experiment,Guizhou Camellia oliefera the polyphenol content for (14.9±3.85mg/g), the polyphe nol content of Zhejiang Camellia chekangoleosa (11.3±4.27mg/g), Camellia yu-hsiensis polyphenol content (15.6±2.40 mg/g), Zhuzhou Camellia olief-era polyphenol content (19.02±1.99 mg/g)Through the static adsorption and desorption experiments, D-3520 resin was selected from 5 kinds of macroporous resins as a suitable material to purify the polyhenols from seeds of camellia oliefera. And then the dynamic properties of adsorption and desorption of the resin were studied. The results showed that the best conditions for the dynamic adsorption pH 4.0,1:4 for bed volume of resin to volume of adsorbing solution, and 4 BV/h of dynamic flow, and the best conditions of the desorption are 60% of ethanol concentration and 1.5:1 for the amount of ethanol to resin bed volume.Methanol was used as solvent to extract polyphenols from seeds of Camellia oliefera, and the antioxidant activity of polyphenols of camellia seeds extract was identified. The results indicated that polyphenols from seeds of camellia oliefera had remarkable antioxidant activity. Monoglyceride was a kind of ideal emulsifier in polyphenols and camellia oil mixture, and it could improve antioxidant activity of polyphenols. Antioxidant activity of polyphenols could be improved with more addition of extract. Antioxidant activity of polyphenols was not as good as BHT, But it was better than BHT when polyphenols was used with BHT. In the range of test concentration,the maximum scavenging rates of polyphenol extracts from seeds of camellia oliefera to hydroxyl radical by Fenton reaction,superoxide anion radical generated by pyrogallol auto-oxidantion were 80.13% and 70.45%, respectively.
