Camellia seed after extracting oil will produce a lot of shell residues. The comprehensive utilization of these camellia seed shells will avoid waste pollution, but also improve the economic benefits of camellia.In this paper, we studied on the evaluation of antioxidant activity of camellia seed shell extracts, polyphenol from camellia shell and qualitative analysis of antioxidant substance.At the beginning of this paper, camellia seed shell was sequentially extracted with ethyl acetate, acetone, methanol and water. The antioxidant abilities of the extracts from camellia seed shell obtained were evaluated by Folin-Ciocalteu, NaNO2-Al(NO3)3, ABTS radical scavenging and DPPH radical scavenging. Results showed that, total solid substance contents were higher in high polar solvents. WE had the highest percentage, but polyphonel content and flavonoid content were low. The methanol extract and the acetone extract had higher polyphonel contents, which respectively were 47.66%,41.97%. The ethyl acetate extract had the highest flavonoid content, which was 72.17%, twice than acetone extract and methanol extract. And it had the strongest antioxidant activity in the ABTS scavenging assay and the DPPH scavenging assay, whose IC50 were 36.2μg/mL,180.3μg/mL All the extracts exhibited strong antioxidant abilities in a dose-dependent manner.Meanwhile, correlation analysis suggested that the antioxidant abilities of these extracts were correlated with the polyphonel content and the flavonoid content. The flavonoid substance made the more important role in the scavenging radicals.And then, choose the suitable solvent to extract polyphenol from camellia shell. Single factor analysis and response surface analysis were employed to optimize extracting parameters including liquid-solid ratio, ethanol weight percent and extraction time. Results showed that the optimal extraction time was for 110 min using a mixture of 54% ethanol/camellia shell ratio (35:1) at home temperature, and the an actual value of camellia shell polyphenol content was 12.2167 mg/g.Finally, camellia seed shell extracts were qualitatively analyzed with the color reaction, TLC and HPLC. The result of the color reaction suggested that the four different solvent extracts all had polyphenol and flavonoid. The methanol extract was isolated with TLC as three spots, neither gallic acid nor rutin, but belongs to flavonoid. The further analysis of HPLC suggested that, the ethyl acetate extract had higher content of rutin. The acetone extract had gallic acid and rutin and gallic acid content was higher. The substance of the methanol extract was complex and the important substances were coumarin and rutin. The water extract also had gallic acid and coumarin. And the ethanol-water of extract had gallic acid, coumarin and rutin.Through the above study, we knew that camellia seed shell extracts had the strong antioxidant ability, and its polyphenol substance and flavonoid substance were abudant. Extracting antioxidant substance from camellia seed shell is a feasible way to utilize of camellia.
