Coordination chemistry of macrocyclic ligands has been a fascinating area of current research interest to the inorganic chemists all over the world. The continued interest and quest in designing new macrocyclic ligands stem mainly from their use as models for protein-metal binding sites in a substantial array of metalloproteins in biological systems, as synthetic ionophores, as models to study the magnetic exchange phenomena, as therapeutic reagents in chelate therapy for the treatment of metal intoxication, as cyclic antibiotics that owe their antibiotic actions to specific metal complexation, to study the guest-host interactions, and in catalysis.Recognition of the importance of complexes containing macrocyclic ligands has led to a considerable effort being invested in developing kinds of macrocyclic compounds.Macrocyclic amide compounds which contain cyclic peptide structure are macrocyclic polyamine ligands. As the unique structure for a class of macrocyclic polyamine compounds, both have the dual structure characteristics of the macrocyclic polyamines and oligopeptides. Amide groups which can be used as hydrogen donors, are also as hydrogen-bonding receptors. The amide through the formation of hydrogen bond generated the properties of interaction between the subject and the object, making the macrocyclic amide compounds with the structure of cyclic peptide as useful ligands in complexing and detecting the harmful environmental and medical anions. Some transition metal complexes with special catalytic activities and biological function, has been paid much attention, and has been widely studied on oxidation, metal enzyme models, molecular recognition and materials chemistry.Macrocyclic compounds occupy an extremely important position in today's sci-ence, Macrocyclic amide compounds' amide bond structure is similar to polypeptide, so whether in organic chemistry or in the life sciences it is a unique advantage.The paper mainly involves the following four parts:1.They made from pyrocatechol with resorcinol,and synthesised the compounds having two nitro-groups which have the chlorine or not,then two nitro-groups are deoxidized two amino-groups through the reduction reaction.next, Macrocyclic amide compounds which made form m-phthaloyl chloride and diamine through condens-ation reaction.2.Three unstable macrocyclic amide compounds were synthesised,and charact-erized by MS.3.Five macrocyclic amide compounds were synthesised.The macrocyclic amide compounds and intermediates characterized by UV, IR,1H NMR and MS.4.We studied the anti-tumor activity test and the recognition properties of nine amino acids.of the stable macrocyclic amide compounds.
