Protein adsorption on the surface often affect the use of biological materials performance, so surface modification of the material which makes it has resistance to protein adsorption performance, and become an important research topic in the field of separation and analysis of the material, therefore the capillary resistance protein adsorption coating has been widely used. Current, the method which improve the capillary resistance protein adsorption on the inner wall is the most commonly used to suppress Si-OH, thus inhibiting capillary for protein adsorption, improve the efficiency of protein separation and separation repeatability. Polymer coating layer mainly divided into physical adsorption coating and chemical adsorption coating, the physical adsorption coating preparation process is supplely, and the stability of the coating is worse that fall off easily; However, the stability of chemical adsorption coating is excellent, but the preparation process is relatively complex and controllability. Based on the above issues, in order to simplify the covalent bonds of coating preparation process and improve the controllability of the coating, we carried out the work as follow:1. Double side with diazo group of photosensitive polyethylene glycol(diazo-PEG)polymer was synthesized, after UV irradiation, the photo-sensitive diazo groups and hydroxyl silicon on the capillary wall light crosslinking curing reaction, the ionic bond convert into covalent bond, thus gaining protein adsorption resistance of capillary covalent bond coating column. The study on electrophoresis coating modified capillary column, effectively inhibit the EOF and impedance of protein adsorption performance, and has good chemical stability and the reproducibility at the same time.2. The photosensitive diazo group of amphiphilic poly(vinyl alcohol)-b-polystyrene polymer(diazo-P(VA-St)) was synthesized by the method of atom transfer radical polymerization. After UV irradiation, base on the self-assembly principle on the inner wall of the capillary, construct the diazo-P(VA-b-St) covalent bonds of coating, and then use the coating for capillary electrophoresis separation of proteins, the experimental results show that the coating has good stability and reproducibility, and can effective impedance protein adsorption.3. The synthesis of thermo-sensitive polyethylene glycol(PEGDA-DTT-dithiols),then using the polymer to prepare the diazoresin(DR)/thermosensitive polyethylene glycol(PEGDA-DTT-dithiols) coating, after UV irradiation, the hydrogen bond can be converted into covalent bonds, then a self-cleaning capacity and has good resistance to protein adsorption performance and stability covalently coating column was fabricated.
