The molecularly imprinted monolithic column have potential applications in HPLC, due to its the advantages of simple preparation procedures, good reproducibility, high selectivity, low backpressure of column and low-consumption of template molecules。This thesis mainly focuses on the preparation of MIP monolithic column in a conventional chromatographic column for Fluoroquinolones separation by high performance liquid chromatography. The synthesis methods of molecularly imprinted monolithic columns were developed for improving the selectivity and contractibility by copolymerization and thermal-initiated polymerization in a traditional stainless-steel chromatographic column using methylacrylic acid (MAA) and methyl methacrylate (MMA) as functional monomer. The effects of polymerization conditions and chromatographic conditions of the imprinted monolithic columns were investigated. The present monolithic MIP was successfully applied for the quantitative determination of ciprofloxacin and enrofloxacin in eggs. The thesis includes three chapters as following.Chapter one:The principle of MIT, polymerization methods, application of this technique and its future developments were introduced. The monolithic column has received increasing attention over the past decade as an attractive alternative for the analysis of complex sample, particularly in complex chiral, environmental and biological samples.Chapter two:A molecularly imprinted HPLC monolithic column for enrofloxacin was prepared by an in situ thermal-initiated. The effect of polymerization conditions and characteristics of the MIPs such as binding capacity, morphological characteristics and thermodynamic were evaluated. Furthermore, Some chromatographic conditions such as mobile phase composition, flow rate, on the retention times and the content of acetic acid were investigated. A rapid separation of enrofloxacin and ofloxacin was obtained under optimizing conditions.Chapter three:The monolithic MIP columns were designed and prepared for rapid separation of a homologous series of quinolones derivatives, ciprofloxacin, and enrofloxacin, by in situ thermal-initiated copolymerization technique. The influences of different monomers and the content of copolymerization monomer on selectivity, contractibility, column pressure of the monolithic column, and chromatography characteristics were investigated. The present monolithic column was successfully applied for the quantitative determination of ciprofloxacin and enrofloxacin in eggs with satisfactory results.
