Validamycin, which is used as one of agricultural antibiotics for rice sheath blight,has been researched and developed for forty years in China. As the varieties of rice sheath blight busters, validamycin has been extended to the wheat, maize, cotton, rape, vegetables and fruit trees and other crops,and even extended to the treatment of diseases of Chinese carps. The market of validamycin products is extending and expandingat present. It is the most inexpensive biologicalpesticide, and is irreplaceable for the farmers in using pesticides.At present, in the production of validamycin, the fermentation titer is the bottle neck. In the first part of this research,natural breeding of high yield strain is used for the screening and breeding a new strain which can yield higher validamycin and morestability. The obtained of high-yield strains is helpful to improve the level of industrialization of validamycin fermentation, reducing production cost of single pot, and will further enhance the validamycin’s market competitiveness.Second, an orthogonal test method is used to find out the influence of carbon source on the production of validamycin in shake flask scale and the fermentation tank scale. The results is as follows: The 7% Rice not only enhance the fermentation level, but also reduce the production cost of validamycin. The optimization fermentation condtions for validamycin is : 3% race, 2% Peanut, 1% yeast, 1% Silkworm chrysalis, 0.2% NaCl,0.0625% KH2PO4,0.2% CaCO3, 0.03% Defoaming agent, pH=7.5, seed age 22-24 hours. In the fermenter medium components as follows: 7% rice,1% peanut, 2% yeast, 1% amino acid, ventilation fermentor 1: 1 V/ V· min, the yield of validamycin is reached 24981μg/mL.Thirdly, through the research of the feeding type, dosage and timing of inspection, repair materials on the yield ofvalidamycin, the optimzation fermentation condition is obtained and the yield of validamycin is promoted(26060 μg/mL). In the fed batch system, 2% liquefied starch sugar is added when the the amount of residual sugar 3.5%.the dissolved oxygen control in the late fermentation process has no effect on the yield ofvalidamycin. This control can slow down the power consumption, you can save electricity costs.Finally, through the analysis of peptide molecular structure and traits of validamycin, improved macroporous crosslinked ion exchange adsorbent is used to separation and purification thevalidamycin.
