We extract genome DNA by the method which so-called minim wholeporcine blood in this experiment. Then, we determine the alleles and genotype frequencies ofHalothane gene and FUT1 gene (two alpha (1,2)-fucosyltransferase gene) in three groupswine, which include two species lines and one commercial line by PCR-RFLP method.Among of them, one group is Landrace origin (Daguanshan group), another is purebredLandrace, purebred large white (Campus farm group), the commercial line is the crossbred ofLandrace and the large white (Shaoxing group). At the same time, we constfuct two fixedeffect models in order to study on Halothane locus and FUT1 locus and analyze their effectsfor the swine performance. By the SPSS software, we analyze the effect of Halothane andFUT1 locus on some traitS of pigs, and analyze the relationship between FUT1gene and thepiglet's morbidity by independence X~2 test.The result indicates that three genotype frequencies of the Halothane gene, which isonly determined in two samples (Daguanshan group and campus farm group), theirfrequencies of three genotypes are 74.3%(NN), 24.5%(Nn) and 1.2%(nn). The two allelicfrequencies are 86.5%(N) and 13.5%(n). In the sample of Daguanshan group, the frequenciesof three genotypes are respectively 75.9%(NN), 23%(Nn) and 1.1% (nn), while those ofallele are 87.4%(N) and 12.6%(n). In another sample, campus farm group, the frequencies ofthree genotypes are 67.3%(NN), 31%(Nn) and 1.7%(nn), whiIe its corresponding aIIelicfrequencies are 82.8%(N) and 17.2%(n).As for FUT1 Iocus in three samples are as follow: 81.4%(GG), 16.6%(AG), and2%(AA). The aIleIic frequencies are 89.7%(G) and 10.3%(A) respectively. In Daguangshangroup, three genotypic frequencies of FUT1 locus are 91.3%(GG), 8.7(AG) and 0%(AA)respectively and the allelic frequencies are 95.7%(G) and 4.3%(A). In campus farm gtoup,three genotypic frequencies are 34.5%(GG), 53.4%(AG), and 12.1%(AA) respectively. Itscorresponding allelic frequencies are 61.2% (G ) and 38.8%(A). In the third sample, Shaoxingcommercial group, the genotypic frequencies of FUTl locus are 84.4%(GG), l5.6%(AG),and 0%(AA) respectively. ltS aIlelic frequencies are 92.2o/o(G) and 7.8%(A).The post weaning pigIets diarrhea morbidity analyzing shows that the morbidity is notrelevant to the traditional Chinese medicinaI compound 'Zhilishensan' which piglet is fedthen, but relevant tO inherit6d susceptibility gene G ofpiglet.We study the effect of the HaIothane and the FUTl gene for tl1e performance of swine.The resultS show that 'N' gene has a significantly effect (P<0.05) on the total number bom inlitteF, 20d survival number and the groWth-rate before or after weaning in pigIet. 'n' gene hasa significantly effect (P<0.05) on the backfat. A dose effect-origin from 'G' gene isdeveloped on the average weight of pigIets bom, the average weight of littCrs, and thesignificantly effect on the groWth-rate after weaning (P<0.05). 'A' gene seems to in favors ofthe fetus surviving. When we investigate the intOracting effect of tWo loci, we get the resultthat the genotype NN-AG composing of dominant homozygote on Halothane locus andheterorygote on FUTl locus has a significant1y complementny effect on the number born inlitter (P<0.05). The genotype Nn-AG, which are hetCrorygotes on two Ioci, possess additiveeffect, comparing to the genotype Nn-GG on the totaI number born in Iitter and the numberborn alive, the former taking on additive effect is l.04 piglets (P<0.05) and l.89 piglets(P<0.01) respectively...
