Studies On The Ways Of Regeneration From Leaf Pieces In Vitro Of Prunus Cerasus

In this paper, the ways of regeneration from leaf pieces of Jingyin No. 1 in vitro were researched mainly in two aspects0 The shoot induction from leaf pieces was one way and the root induction was the other way0 The latter way was studied preliminarily and leaf pieces was infected by Agrobacterium tumefaciens, then kanamycin callus and roots were obtained0The results showed: on WPM-based medium with 2mg/i BA~ 2mg/i IAA by using furled leaves below the shoot tip after3 weeks, the shoot differentiation frequency reached 96. 0% with 9.40 mean number of shoots per regenerating explant0 The basic medium explant types and exogenous hormone were the key factors0 The roots would appear from the leaf pieces after 2 weeks on F14-based medium with 5mg/i IBA. the root differentiation frequency reached 100. 0% with 8. 52 mean number of roots per leaf pieces0 The dark treatment.. exogenous hormone were the key factors0 150mg/i cefotaxime was able to inhibit the growth of Agrobacterium tumefaciens and there was no significant effect on the roots differentiation0 30mg/i kanamycin was able to inhibit the roots differentiation if the leaf pieces was cultured for 3 days on medium without selectable antibiotic. And lOOI.tmol/l AS could improve the transformation, the time of dipping in the Agrobacterium tumefaciens had no obvious effects.
