Avian Paramyxovirus Type I isolate YG97 from goose and isolate Y98 from chicken have been continiously propgated in chicken embryo fibroblast (CEF) of 9-day- old SPF chicken for 8 generations .The infected cell for every passage were observed with optical microscope .The results were as follows:There is no obvious cytopathic effect (CPE) from first passage to eighth one ,but in subgeneration of CEF having been infected for 60-72 hour ,there is CPE .By means of immunofluorescence technique ,we know both isolate YG ~ and isolate Y98 replicate in cytoplasma conforming to paramyxoviridae general characteristics.Ultrathin sections of infected CEF were observed with transmission electron microscope . The results suggested that the two new isolates can cause organelle of CEF gentle ultrastructural pathogenesis. Above results suggested that there was differentiation in characteristics of cell culture between isolate YG97. isolate Y9~ and Newcastle Disease Virus strain F48E8.With transmission v electron microscope ,the morphological processing of the two new isolates in CEF has been observed .The are origined from filament-like structure, generated in nucleus and transported to cytoplasma. The influences on various organism cells of SPF chicken artificially infected with isolate YG97 and isolate Y98 were observed with scanning electron microscope and transmission electron microscope. The two can cause ultrastructural pathogenesis widely in many organs and tissues such as hepar, pancreas, lien, ren, pulmo, cerebral, cerebellum, cor, muscular and so on, of them ,caused by i~olates YG97 and Y98 more obvious in epithelial cells of digestive tract and unobvious in epithelial cells of respiratory tract ,but ultrastructural pathogenesis caused by NDV strain F48E8 ,more obvious and more serious in glandularis and muscularis ventriaculi and in epithelial cells of respiratory tract .Above results suggested that different isolates brought with them different affinity to the same tissues ,even the same isolates could brought with them different affinity to different tissues.
