Objective:Microsatellite instability(MSI) has been used to detect widely the genome of tumor human.One of the chicken tumor disease caused by MDV,harm overwhelmingly,but there is reported rarely about MSI in Marek's disease virus which lead to the tumor.The investigation of microsatellite instability of some chromosomes of the chicken embryo fibroblasts infected with the virulent MDV RB1B strain aims to provide basic data for biology of formation and development of Marek's disease in terms of host genome.Method:In this study,chicken embryo fibroblasts were prepared from 21 SPF chicken embryos.The half of CEF derived from each SPF chicken embryo were infected with the virulent MDV-RB1B strain,the others were used as control.Their genomic DNA was extracted,respectively.MSI of some chromosome were detected at 43 microsatellite loci by means of PCR,PAGE electrophoresis and silver staining in matched normal CEF and MDV-infected CEF.Result:MSI were found on 38 of 43 loci,the incidence rate of ABR0262 was 38.10%and that of ABR0048,ABR0041,MCW0183 were all 33.33%,MSI was found on all embryos,the incidence rate of sample 9 were up to 39.53%,and the incidence rate of sample 11 were up to 30.23%.Conclusion:The results showed that MSI was found on some chromosomes of CEF infected with MDV.When infecting CEF,MDV can lead to extensive changes in the host genome.These changes are random,but regular to a certain extent,which can be detected by microsatellite markers.
