With the widespread cultivating and constant emerging in large number of new hybrid rape, the traditional method of identifying the authenticity of the hybrid rape F1 can/ t meet the managerial needs. It is imperative to study the accurate and economical identifying technical methods in production. In this test , we carried out SEM observing, isozyme, protein electrophoresis and RAPD technique on the three-line system hybrid and variety rapeseeds ,which were widely used in production. We drew the conclusions as following: SEM could be severed as a supplementary method of identifying the authenticity of hybrid rape. Hybrid rape F1 manifested many common grounds with three-line system through SEM because of their close relationship. However, there still have fine distinction among them . Esterase isozyme , peroxidase isozyme could be used to appraise and distinguish the authenticity of hybrid rape F1. RAPD signal is an reliable ,accurate identifying technique : Using the modified method, we extracted the genomic DNA samples from rape seedling. Four RAPD markers ( D07 D18 E03 and E19) were chosen from 100 primers. The band pattern of the F1 hybrid contained that of its parents (male sterile line and restore line).The band pattern of maintainer line seeds was much similar to that of male sterile line.RAPD technique is more accurate and reliable than isozyme one .The kinds of isozyme are the products of gene expression , but RAPD technique can report the information of gene .The difference in the level of genomic DNA ,among the hybrid rape F1 and its three-line system , can be identified using RAPD markers .RAPD technique is more reliable ,accurate than isozyme electrophoresis in that it can serve as unambiguous markers in spite of environment. The results will be much better using these two methods simultaneously.
