31 species and hybrids of grape were analyzed by techniques of isozyme markers, and 11 amongst were analyzed by means of RAPD markers and assistant with morphological markers. Different analysis methods showed varied dendrograms, either by the same cluster analysis with two kinds of markers respectively or by each kind of markers with different methods of cluster analysis. The dendrogram from the RAPD markers might be better in reflecting the relationship of the species and hybrids when linked to the morphological markers. It was hard for the isozyme markers to reveal the differences among those with very close relationship since lack of marker sites the single enzyme could afford, and also marks of the sensitive enzyme were influenced by the factors of environments, and sampling time and positions, which would affect the veracity and dependability of the results. With adequate mark sites, the RAPD marker analysis was competent for precise revealing differences and relationships among the species and hybrids of grape.Method for DNA extract in this study was SDS, which was different from the conventional CTAB previously used in grape plants. The productivity and purity with 1.6~1.7 of extracted DNA matched well for the requirement of RAPD-PCR.
