Peach is a kind of fruit trees, which cultivated popularly in the world. Y/y gene and F/f gene are the two most important agricultural characters related to the artifactitious process. So it is important to explore the SCAR marker, which closely linked to the character of Y/y flesh in the canned process and breeding. The RAPD system was optimazed and the PCR system was made more special through our study. Fl population from common peach (Prunus. Persica L) Jingyu and Flavor hybridization were used as the experiment meteral, The RAPD marker OPU03-900 linked to Y/y gene and OP107-1000 linked to F/f gene were successfully converted into specific, stable, and reliable sequence-characterizied amplified region (SCAR), which were named SCU03-900, SC107-1000 respectively.25bp and 27 bp primers were designed according to the sequence we got before, then the SCU-900 SCAR marker was developed by the way of increasing of anneal Tm.When we convert SCI07-1000 SCAR marker, the primers came from the known sequence amplified DNA in both kinds of F/f plants. So we used a new way in which 2 base pairs were added to unknown 5 terminal of the RAPD primer one by one, and at the same time 3 base pairs were added to known 3 terminal of the RAPD primer. Then the RAPD marker OPI07-1000 linked to F/f gene was successfully converted into SCAR marker.In a word the two SCAR markers are very valuable to the assistant selecting in the peach breeding and the artifactitious industry of peach.
