Flue-cured tobacco with special aromas is a new kind of variety developed in our country, it possesses the similar aroma character in oriental tobacco. Great attentions have been payed to its utilization both in leaf tobacco production and cigarette industry. In order to find out its aroma compositions and the genetic characters of its aroma trait, in this research , for the first time, the main aroma and flavor constituents in it have been identified and analysed; The genetic analysis of the special aroma trait have been done; And an optimalized tobacco RAPD reaction system have been set up, by using this system, we have attempted to find the RAPD marker of the special aroma property, the main results are as following.1. There are great differences between flue-cured tobacco with special aromas and Oriental tobacco in terms of constituents, while there are relatively small differences between two types of flue-cured tobacco. Flue-cured tobacco with special aromas has the special components of l-penten-3-ol, 1-pentanol, 3-hydroxy-2-butanone; and has five kind of constituents of 2-pentenal, nonanoic acid et. al., which are also determined in Oriental tobacco; According to the constituents determined in three types of tobacco, 2,4-heptadienal, benzaldehyde, benzeneacetaldehyde, 4-[2, 6, 6-trimethyl]-3-buten-l-one, 1~[2, 6, 6-trimethyl]-2-buten-l-one are most abundant in flue-cured tobacco with special flavoring aroma.2. The special aroma character is regarded dominant heredity as principle, the quality of the special aroma is controlled by one single dominant gene, the amount of the special aroma is probably controlled by incomplete dominant gene or polygenes.3. An optimalized RAPD reaction system which is suitable to tobacco was set up. By employing this system, RAPD polymorphic marker of two flue-cured tobacco parents with contrary aroma property were studied. The polymorphic fragments that amplified from the four primers, Y233, Y466, Y102, Y386, which were selected from 500 random primers could be used as molecular marker ofthe two tobacco varieties.By employing the polymorphic fragments that amplified from primer Y466, the heredity of aroma property in two flue-cured tobacco parents,the Fl.and F2 were tracked. Elementary analysis showed that the polymorphic fragment has the trend of genetic linking with the aroma property , and may be the molecular marker that link with special aroma trait.
