On the base of morphological character and isozyme analysis, making use of RAPD and ISSR to analyze heredity backgrounds of 30 germplasmic resources of chewing cane, selecting some primers with amplifying clearly and better polymorphic degree and stable dependable (Primer SS and WW can accurately examine the differentiation of Badila, local chewing cane and hybridized varieties), establishing a set of heredity background data from character-protein-gene, for providing the theories the basis on the germplasmic resources of chewing cane in the identification and evaluation, heredity classification, fingerprint diagram etc, and also will further increase the level of the study and use of the germplasmic resources of chewing cane. The results are as follows:1. Morphologic characteristic: The local chewing canes of our country have the basic characteristic of Saccharum officinarum L. with big stem, soft skin, much juice and broad-bigness leaf. The color of its stern epidermis is all regarding green as the main colour, but also there is part germplasm to report light purple or brown, and they contain obvious difference with dark purple of Badila. The shape of internodes, primarily there is cylindrical form, barrel form,spindle form and obconoidal form. They mostly infect sugarcane mosaic virus disease.2. Esterase i sozyme: The results indicate that esterase isozyme of the local chewing cane is not serious for variation degree and not evident for variation the boundary. There are totally present 8 enzyme bands:2A,4A,6 A,8A,10A, 12A, 14A and 20A.There are 4 kinds of zymogram type: Zymogram I (6,8,10)A, zymo-gram II (2,4,6,8,10)A, zymogram III (2,4,6,8,10,12,14)A and zymogram IV (2,4,6,8,10,20)A. Zymogram takes the boundary clear. Among them present 36 varieties of having zymogram II that shares the germplasm of chewing cane to amount of 87.7%. Zymogram I only has Fengcheng purple chewing cane and Badila. There are 3 varieties presenting zymogramlll or IV , they are respectively We-nling chewing cane, Leizhou chewing cane and Baishan sugarcane .3. RAPD ana I ys i S: On the foundation of the optimum RAPD respond system and respond procedure, acquiring the heredity polymorphic data of chewing cane germplasm, 30 RAPD primers totally amplify to out 445 bands, with an average of 14.8 bands per primer. Among varieties have 253 polymorphic bands that occupy 56.8% of the total number. The results of unweighted pair group method with arithmetic means analysis(UPGMA) show that similar coefficients of chewing2cane are all at 80% above. Genetic relationship among germplasm is high similarity, but still we can see some differences. At the level of D=0.86, the chewing cane germplsems are separated into 5 types: The first, second types with Fujian province chewing cane are dominant. Category 3 is outside of Fujian province chewing cane for dominance. Category 4 is dominance for two first steps making sure for hybrid. Category 5 is confounding, there is outside and inside of Fujian province chewing cane.4. ISSR ana I ys i S: On the foundation of the optimum ISSR respond system and respond procedure, acquiring the heredity polymorphic data of chewing cane germplasms, 14 ISSR primers totally amplify to out 168 bands, with an average of 12.0 bands per primer. Polymorphic bands take the comparison to 61.35%. UPGMA results show that similar coefficients of chewing cane are mostly at 70% above. Genetic relationships among germplasm are high similarity. At the level of D=0.76, the chewing cane germplsems are separated into 3 types: The Category 1 primarily is the chewing cane germplasm in the Fujian province. Category 3 is outside of Fujian province chewing cane for dominance. Category 2 is confounding, there is outside and inside of Fujian province chewing cane.This issue of gene set DNA extraction, PCR technique system, genetic diversities, classification, conspecific or hetero-named phenomenon etc. are discussed.
