Thirty-eight ramie varieties including 30 wild species and 8 cultivated species wereused to analyse the genetic multiformity and correlation by RAPD, ISSR markers andisoenzyme technic. The results are as follows:1. The results of esterase isoenzyme analysis indicated that total 146 strips ofesterase isoenzyme were detected in 38 varieties. Seven strips to nine strips were detectedin cultivated species, while only 1 to 5 strips in wild species. The ratio of the strip movedRf was 0.381 to 0.835. Cluster analysis indicated 38 varieties were classified in 5 clusterswhen R=0.79.2. Utilized B.nivea and B.clidemiOides as DNA tamplate, which had great differencerespectively, to optimize amplification system by double orthogonal design, which hadfounded the RAPD-PCR optimization of amplification system for Boehmeria plant. Theoptimized RAPD reaction system include: total mixture 25ul, RAPD primer 0.75umol/L,sample DNA 150 ng, Mg2+ 2.1 mM, dNDP 0.2 umol/L, Taq DNA polymerase 1.5U. Theoptimized RAPD reaction programmes was: 94℃for 4 min, and then 36 cycles including94℃for 45 sec, 37℃for 45 sec, 72℃90 sec, with a final elongation step of 10 min at 72℃and 4℃preservation.3. Through the analysis of 38 varieties genetic multiformity and correlation byRAPD, ISSR markers, the result showed 358 strips were amplified by 31 RAPD primerswhile 266 strip by 18 ISSR primers. Each RAPD and ISSR primer amplified 11.5, 14.8 onaverage. Clustering analysis by NTSYS software indicated 38 varieties were clustered 8clusters when supposed R=0.73. The result has a significant relativity with the varieties'area.4. Through the analysis of RAPD and ISSR molecular markers by NTSYS andcomparasion with compositive result, we could conclude that RAPD and ISSR were allsuited to analysis genetic multiformity and correlation. These result had a significantrelativity between them.
