In this research, by using 18 varied strains of Pyrus Brestchneidery Rehd cv Jinhua as experimental materials and Pyrus Brestchneideri Nak cv Jinhua as CK, the researcher observes and analyzes Jinhua's time of phenology, features of its leaf and fruit, medical index, photosynthesis efficiency of its leaves and economic characters of its fruit were observed and analyzed in the experiment. DNAs from 18 varied strains of Jinhua and the leaves of 2 Pyrus Brestchneideri Nak cv Jinhua and makes and analysis of AFLP molecular marker were mode at the same time. The results showed that:1. Jinhua is apt to vary geneticly in the long term cultivation process. Genetic variation appears in 18 varied strains of jinhua and clustering analysis of AFLP showed that: (l)when genetic similarity coefficient is 0.67. J1, J2,J6,J3,J4,J5,J7,J9,J10,J8 as a category; J11, CK1,CK2, J13,J14, J15,J16,J17,J18,J12 as a category. (2) when genetic similarity coefficient is 0.91. The results of UPGMA showed that the 18 varied strains were divided 10 categories. They were J1, J2,J6,J3,J4,J5,J7;J9,J10;J8;J11;CK1,CK2; J13. J14,J15;J16,J17,J18;J12.2. The analysis of Venturia pirina Anderh medical index of 18 varied strains leaves indicates the disease-resistance of each varied strain: J4 is the strongest then followed by J17,J15,J7,J6, J3,J8. The ability resist Venturia pirina Anderh is decided by heredity of each strain. The high moisture and 20-24癈 temperature are the most favorable conditions for the disease.3. The analysis of medical index of Gymno sporangium haraeanum Syd of Jinhua varied strains shows the disease-resistance of each strain: the strongest are J16,J3,J15,J18,J13,J1,J7, then followed by J5, J10,J4,J2,Jl7,J6,CK. Such disease-resistance is determined by heredity of each strain and is also closely related to the host in the envioroment, moisture, wind.4. The analysis of medical index of the fruit 18 varied strains of Jinhua indicates: the strongest areJ16,J13,J8,J3,J5, followed by J7,J14.5. The ananasis of photosynthesis efficiency of leaves shows that the photosynthesis efficiency of Jinhua varied strains are all higher than P. pyrifolia Nakai. Nanguo and Changxi and the difference between each strain is decided by illumination.6. The research into ways of extraction of Genomic DNA of Jinhua with high quality and high purity indicates that improved CTAB is the best way to extract DNA of pears, that the appropriate34quantity of extracting liquid 1g fresh material is 5ml and that the critical time for immersion extraction under 65℃ is 30' and keeping 65℃ constant temperature for an hour is the most favourable conditions for extracting high-quality and high-purity DNA needed by AFLP. Under such conditions, DNA can get high yields.7. The analysis of AFLP of Jinhua varied strains shows that:(1) Only the complete and thorough dual enzyme cleavage can lead to steady amplified bands; the molecular marker of AFLP requires large fragments of moldle plate DNA, no decomposition reaction, no RNA, 1.7-1.9 OD(260)/OD(280).(2) The accurate design and preparation of primers is the prerequisite to obtain numerous amplified sites, high polymorphic, high quality and high identified rate.(3) ECORI/MseI is very important Internal cutting enzyme of Jinhua varied strains and among selective bases, ECORI-ACC/MseI CAT. That is the selective bases which require more AC are more suitable to the expansion of Jinhua varied strains.(4) The analysiss of Jinhua varied strains indicates: a pair of primers expand 17.7 sites on the average, amony which polymorphic sites are 11.3 on the average, which holds a high proportion of 66.9% and has a genetic diversity.(5) The feature bands of AFLP fingerprint atlas can be used as the marker of far and near kinship and character variation, which provides basis for breeding, recording new breed and shifting to appraise gene of destination and also supplies adjective, scientific and accurate technical appraising for the...
