Endosperm of loquat was used for explants in the study on in vitro culture and plant regeneration in "DaWuXing" loquat. The main subjects of the study were callus induction N plantlet regeneration N embroid induction xplantlet rooting and transplanting of rooted plantlets. The results were as follows:l)The best way of material sterilizing was as follows: fruitlets-removal of sepals and hair-washing 1 hr with tap water- immerseing in 75% alcohol for 10m-washing once with sterile water-seed immersed in 0.1 % mercuricchloride solution for 4m.2)The suitable time of taking endosperm for culture was about 7w after pollination when endosperm was in free-cell phase.3)A low temperature treatment of the fruitlets for 17 days could result in a rate of 35% in callus induction. A seed pre-culture for 2-4days on MS added with GA3lmg/K 2,4-D0.lmg/K 6-BA5mg/l could also remarkably improve the frequency of endosperm callus occurrence.4)Endosperm culture with embryo could remarkably improve the frequency of callus induction and GA3 could partly replace the role of embryo.5)Cultured on MS+6-BA2mg/l+2,4-D0.5 ~ 0.7mg/l+GA30.4mg/l, the frequency of endosperm callus induction was as high as 33.3%.6)The smaller size (0.15g? of callus of the first generation multiplied better on MS+6-BA2mg/l+2,4-0.5mg/l.7)Callus cultured on MS+Ze2~3mg/l+NAA0.1mg/l for 30 days, then transfered to the same medium added with 0.3mg/16-BA could result in a better shoot differentiation. The rate of shoot multiplication could reach 3.2 when cultured on MS+6-BA0.5mg/l+KT0.5mg/l +IBA0.02mg/l+NAA0.02mg/l.8) High concentration of 2,4-D combined with certain GA, concentration benefited embroid induction.9)Strong shoots cultured on l/2MS+IBA0.25mg/l+NAA0.25mg/l+AC0.2% could give a rooting rate of as high as 80%.10)The transplanting survival rate of rooted plantlets after 5~7days of hardening could reach as high as 80%.
