Twenty samples of Beiwuweizi collected from Heilongjiagn, Jilin, Liaoning, Beijing, and Nanwuweiz brought from Sichuan, Hunan, Shanxi and Xian were analysed by RAPD and ISSR molecular markers. Twenty RAPD primers and 9 ISSR primers were selected from 100 and 30 respectively, and could produced qualified fragments. Among Ihese selected primers only 8 RAPD and 5 ISSR primers could identify the two different Wuweizi. DNA fingerprints were obtained by PCR with all selected primers and analyzed using cluster analysis. The fingerprints indicated that amplified fragments of the same variety were almost the same, while only few primers could get the specific fragments able to identify the two species. The genetic similarity of the two varieties were 0.575. The genetic similarity of Nanwuweizi and Beiwuweizi of each variety ranged between 0.856 to 0.947and 0.902 to 0.993, respectively. These figures provided the same information as that of fingerprints, that is the genetic diversity inside varieties were tiny, and relatively big between them.Content of Schizandrin, the mainly effective compound was determined by high performance liquid chromatography, the method used in People's Republic of China pharmacopeial convention. Also, the high performance capillary electrophoresis was used. The figures got with the two methods were very similar and r=0.9596. The content of Schizandrin in all the Beiwuweizi were above 0.4%, which is the standard in pharmacopeial convention. Thus, all the Beiwuweizi we detected were of good quality. Nancha produced the best quality Beiwuweizi, well Benxi and Huma were next. In concern of the DNA fingerprints, primer SBS D18 amplified a specific fragment within wuweizi produced from the three areas, while not in other samples.The selected primers could be used in identifying Nanwuweizi and Beiwuweizi. Nancha produced the best quality Beiwuweizi containing highest content of Schizandrin, and should be considered first while culturing new good variety. The specific fragment amplified by primer SBS D18 could be used as a potential DNA marker indicating good quality of wuweizi, especially useful in the use of identifying good seedling in an early stage of growth.
