The purpose of the study was to test the possibility of obtaining somatic hybrids by means of shoot-tip grafting in vitro. The trial materials including Poncirus trifoiiata and C.reticulata and C.grams were used in the study. After some weeks callus and adventious buds from grafting position were cultured in vitro. The resultant plantlets were appreciated finally.The results were as follows:(1) Suitable stocks and scions were from the seedlings cultured in the dark for two weeks or so.(2) The highest frequency of shoot-tip grafting was obtained by means of chair grafting, however, in consideration of the whole process, apical grafting was most suitable than others.(3) All the treatment could enhance the successful frequency of shoot-tip grafting and weight of callus from grafting position and the sprouting frequencies of buds from grafting position.(4) For primary culture of callus, the optimum multiplication culture medium was MS+0.5mg/L BA+0.5mg/L IBA+0.2mg/L NAA+suger30%.(5) The optimum medium for redifferentiation callus was MS+0.5mg/L BA +1.0mg/L KT+0.1mg/L IBA +suger30%.(6) The optimum medium for the multiplication of adventious buds was MS+0.5mg/L BA+0.2mg/L KT+0.1mg/L NAA+suger30%. In which multiplication , coefficient could reach 11.5 in 60d.(7)By means of morphological identification ,plantlets with characters of both stock and scion were obtained.
