This research was to study the effects of various electrical activation protocols in rabbits oocytes, in order to cross reference to the study of "Rabbit oocyte cytoplasm supports development of embryos produced by nuclear transfer of human skin fibroblast cell."In experiment 1. three different electrical activation procedures were studied: (1) 2 pulses, double-click, 640 usec,0.2kv/cm, (2) 8 pulses, 12min apart, 40usec,0.6kv/cm, (3) 2 pulses, 5sec apart, 20usec, 1.3kv/cm. There is no obvious difference between the activation efficiency of treatment 2 (88.9%) and treatment 3 (73.9%) . The activation efficiency of treatment 1 was much lower (50%). As to the efficiency of development to Scells and further, treatment 3 (63.0%) is remarkably higher than the others (10% and 8.3%, respectively). 43.5% of the embrys in group 3 develop to the morula stage and 6 of them reached the blastocyst stage.In experiment 2. we investigated the difference of the oocytes acquired from oviduct and ovary. Both the percentage of activation and development, the oocytes from oviduct was significantly higher than those from ovary.The third experiment is about the concentration of the pulsating medium. The results indicated that much more oocytes developed to 8 cells in 0.25Mmedium(p<0.01).
