The research on technology of In Vitro maturation, parthenogenesis and In Vitro fertilization of bovine oocytes plays an important role in In Vitro production and application of embryo. In this article, we use ethanol (EN) and ionomycin to activate matured oocytes, use Ca2+ fluorescent probe Fluo-3/AM to observe the spatial distribution of freeing Ca2+ in cytoplasm of oocytes, and use TCM199 as basal medium to investigate the effect of EGF in bovine oocytes IVF and cleavage and its expression in parthenogenetic embryo and IVF embryo. The results are as follows:1. Cleavage rates and blastocyst rates were highest in the treatment of 7%EN 5min, 2 mmol/L 6-DMAP 4h. Cleavage rates were highest in the treatment 7μmol/L Ionomycin 5 min, 2 mmol/L 6-DMAP 4h, and the highest blastocyst rates were found in the treatment of 10μmol/L Ionomycin.2. Oocytes cultured in mature solution which adds 50 ng/mL EGF certificate highest maturing rates and cleavage rates.3. EGFR was detected expression in the surface of oocyte, 2-cell, 4-cell, 8-cell, 16-cell embryo of IVF and in the surface of 2-cell, 4-cell, 8-cell ,16-cell embryo of parthenogenesis.
