Since Australian scholars Wilkins and Williams first introduced the term of proteome in 1994., proteomics became one of the focus of life scientists. For the nearly changeless of genome, these problems, that the expressing time, expressing quantity of gene, decoration of protein post-translation, and their sub-cell distribution, couldn't be solved simply by gene DNA sequencing. As protein is the expressing production of gene, and proteome is dynamic, space-time, adjusted, which could solve all the above problems. The nature and regulation of life on the integrative level of cell and life organism also could be solved by it. Through the study of data integration of proteome and gene, we can get plenty of data of the functional gene. What's more, the data of proteome can promote the development of borderline subjects like biological informatics, other modern borderline subjects and the medical industry.A fine anther protein IPG-two-dimensional gel electrophoresis (2-DE) technology had been set up by using the material of thermo-sensitive genie male sterile rice of doublelow-s; optimized2-DE conditions of rice anther proteins. Chose good lysis solution, appropriate sample amount and electrophoresis parameters, fine quality 2-DE patterns of rice anther proteins had been obtained by scanning and analyzing through PDQUEST 2D software. Initial ascertained the molecular weight and isoelectric of protein. Studied from the 2D patterns, anther protein spots almost distributed at pH4-8 and molecular from 60KD to 20KD, and most concentrated at pH4-6.7 and molecular 46KD-20KD.The changing of anther proteins of thermo-sensitive genie male sterile rice of doublelow-s and the conventional rice (which is from thermo-sensitive genie sterile rice of doublelow-s) at varies developing times had been studied. The results are as follows:1. Most of the proteins of the conventional rice and doublelow-s at varies developing times were changeless, such as protein spots (70KD, pI5.3), (66KD, pI5.3), (70KD, pI5.5), (66KD, pI5.5 ) , (34KD, pI5.2), ( 34KD, pI6.7 ),( 41KD, pI5.9 ), (41KD, pI6.2), (40KD, pI6.0), which means that they are the housing proteins of rice. From the 2 DE patters of varies developing times of doublelow-s, we found that the protein spots reduced with the development of anther.2. On the conventional rice, protein spot (39KD, pI7.3) was expressing on mono-spore cells, but disappeared on meiosis cells. The expressing quantity of protein spot (14KD,pI5.4) and protein spot (45KD, pI6.5) was larger on mono-spore cells than that on meiosis cells; the expressing quantity of protein spot (pI6.0) and protein spot (10KD, pI4.69) was larger on meiosis cells than that on mono-spore cells.3. Under the condition of fertile temperature of doublelow-s, protein spots (22KD,pI7.8), (22KD, pI6.2), (22KD, pI6.24) were expressing on mother pollen cells, but disappeared on meiosis cells and mono-sporecells; protein spots (p!7.6), (pI7.8) were expressing on mother pollen cells and meiosis cells, but disappeared on mono-spore cells.4. Under the condition of sterile temperature of doublelow-s, protein spot (22KD, pI7.8) was expressing on mother pollen cells, but disappeared on meiosis cells and mono-spore cells.5. Under the deferent temperatures of doublelow-s, there were only expressing quantity deference of some protein spots on mother pollen cells; on meiosis cells, protein spot (pI6.06) only occurred on sterile temperature, but disappeared on fertile temperature;. On mono-spore cells, protein spots(pI7.6) and protein spot (pI7.8) only occurred on sterile temperature, but disappeared on fertile temperature; protein spot (14KD,pI5.4) occurred on fertile temperature and the conventional rice, but disappeared on sterile temperature.Studies on the anther proteins at various development and various temperatures manifested some housing proteins which maintain the basic metabolism of rice; some proteins occurred with the development of pollen, which means that they were the...
