Two-dimensional electrophoresis (2-D) technology had been used broadly to analyze the protein. It included steps such as protein sample preparation, IEF electrophoresis, equation of IPG, SDS-PAGE electrophoresis, silver stain, software analysis etc. In this experiment, TCA-acetone was used to prepare protein sample, IEF and SDS-PAGE electrophoresis were carried out on CELL (BIO-RAD), and the protein maps were analyzed through PDQuest software (BIO-RAD) by VERSER DOC 3000.The results of 2-D showed that there were many differences between the protein maps of the seeds, leaves and anthers of the cytoplasmic-nuclear male sterile line NJCMS1A and those of the maintainer line NJCMS1B. Five spots appeared in the protein map of the seeds of NJCMS1A which didn't exist in that of NJCMS1B, meantime two spots emerged in the protein map of the seeds of NJCMS1B which didn't arise in that of NJCMS1 A. Eight spots were found in the protein map of the leaves of NJCMS1A, which didn't appear in that of NJCMS1B. Forty six spots appeared in the protein map of the anthers of NJCMS1A which didn't emerge in that of NJCMS1B. The above differences might be due to the different cytoplasmic genes, for the male sterile line NJCMS1A and its maintainer line NJCMS1B had the same nucleus but different cytoplasm.Similarly there were some differences between the protein maps of the seeds, leaves and anthers of the cytoplasmic-nuclear male sterile line NJCMS2A and those of the maintainer line NJCMS2B. One spot appeared in the protein map of the seeds of NJCMS2A which didn't exist in that of NJCMS2B; meantime five spots arose in the protein map of the seeds of NJCMS2B which didn't appear in that of NJCMS2A. Twenty five spots appeared in the protein map of the leaves of NJCMS2B which didn't emerge in that of NJCMS2A.
