Now with the improvement of people' s living, white grain wheat hasn' t been to meet people' s need. Black grain wheat is rich in nature black pigment, vitamins, protein, amino acid, and beneficial microelements, especially contents of natural Se resistant to cancer, so it is becoming more and more popular. It is necessary to find high quality wheat with blue or purple grains. Location of the grain pigment gene is the precondition of the assistant wheat breeding.In this experiment, through the genetic analysis of the F3 seeds clour, derived from LUOZHEN 1(purple grain wheat) and 8901(white grain wheat), the result indicate that the colour of grains is controlled by two independent genes. An F2 population was used to map the grain pigment gene by microsatellite and BSA(bulked segregant analysis). Purple-grain and white-grain DNA pools were established using the extreme purple-grain and white-grain plants of F2 population,respectively. Among the 230 pairs of microsatellite primers, two markers were found polymorphism between the two pools. Linkage analysis showed that microsatellite marker Xgwm47 and Xgwml55 were linked with the two purple pigment genes, respectively.Through analysis of Joinmap 1.4 (software), one of the purple pigment gene was located on 2AS and the genetic distance between the SSR marker Xgwm47 and this gene was 2.8 cM, the other purple pigment gene was located on 3AL and with a genic distance of 14.7cM to marker Xgwml55.
