Effect Of Addition Of Cholesterol To Extender On Changes Of Phospholipids And Cholesterol Contents In Cashmere Buck Spermatozoa During Freezing Process

In the study, the effects of addition of cholesterol to the extender on the changes of phospholipid and cholesterol contents in cashmere buck spermatozoa during freezing process were analyzed.The extender containing Tris, citric acid, glucose, yolk and glycerol, was used. After freezing, we achieved the result of 46.99% motile spermatozoa. The highest motility was 58%. Adding cholesterol to the extender did not influence the motility of spermatozoa after frozen-thawing, but increased the rate of acrosome partial-loss and decreased the rate of acrosome total-loss. As the data was not sufficient, furthermore study was required.The pohspholipid contents of spermatozoa were 404.86, 423.74, 383.27 and 333.39nmol/109spermatozoa in semen, post-diluting, post-cooling and post-thawing, respectively. The differences were not significant (P>0.05) among them. The cholesterol contents of spermatozoa were 246.01,190.35,141.91,111.00nmol/109 spermatozoa, respectively. Only the data between post-cooling and post-thawing were similar (P>0.05), the others were significantly different (P<0.05). Adding cholesterol to the extender did not influence the cholesterol contents, but influenced the phospholipid contents. There were no significant differences in C/P ratio among control group, experiment 1 and experiment 2(P>0.05).
