Goat semen cryopreservation technology plays an important role in improving the utilization efficiency of rams,accelerating quality improvement and reducing the risk of transmission diseases.At present,the sperm structure and function of the goat sperm preserved by liquid nitrogen have been changed seriously after thawing,which makes the cryopreservation effect of goat semen not ideal.After freezing and thawing,sperm viability,plasma membrane integrity,and acrosome integrity were not as good as those of fresh spermatozoa.Therefore,it is particularly important to explore techniques to improve the quality of sperm after freezing and thawing.In this study,cashmere goats were used as research objects,and different antioxidants were added to the frozen diluents to study the effects of different antioxidants on the cryopreservation of semen,thus laying a theoretical foundation for improving the cryopreservation effect of cashmere goat semen.The contents are as follows:1.Effect of melatonin on cryopreservation of cashmere goat semen.In order to improve the cryopreservation of cashmere goat semen,different concentrations of melatonin?MLT?were added to the frozen diluent.After thawing,Sperm movement parameters,sperm progressive motility,membrane integrity,acrosome integrity,DAN integrity,mitochondrial membrane potential and reactive oxygen species?ROS?level,and were evaluated by flow cytometry and computer-assisted sperm analysis system?CASAS?.Results,After thawing,semen freezed in the semen extender supplemented 0.5 mg/mL melatonin showed significantly higher sperm progressive motility,sperm,DAN integrity,and mitochondrial membrane potential?52.7%,45.8%,77.9%,95.1%and2.7,respectively?compared to control group?47.8%,36.3%,53.2%,93.5 and 1.9,respectively,P<0.05?.Moreover,ROS level in the sperm was decreased significantly while melatonin was supplemented semen extender.Conclusion,adding 0.5 mg/mL melatonin to cashmere goat semen freezing diluent can significantly improve the cryopreservation effect of cashmere goat semen.2.Effect of coenzyme Q100 on cryopreservation of cashmere goat semen.In order to improve the cryopreservation of cashmere goat semen,different concentrations of coenzyme Q10were added to the frozen diluent.After thawing,Sperm movement parameters,sperm progressive motility,membrane integrity,acrosome integrity,DAN integrity,mitochondrial membrane potential and reactive oxygen species?ROS?level,and were evaluated by flow cytometry and computer-assisted sperm analysis system?CASAS?.Results,After thawing,semen freezed in the semen extender supplemented 400?g/mL Coenzyme Q100 showed significantly higher sperm progressive motility.ROS level and DAN integrity?60.2%,1124 and 93.6%,respectively?.When Coenzyme Q10 concentration is 40?g/mL,sperm membrane integrity,acrosome integrity and mitochondrial membrane potential?63.0%?76.5 and 2.9,respectively?all significantly higher than that of the control group?47.8%,36.3%,53.1%,93.5%and1.2,respectively,P<0.05?.3.Effect of catalase on cryopreservation of cashmere goat semen.In order to improve the cryopreservation of cashmere goat semen,different concentrations of catalase were added to the frozen diluent.After thawing,Sperm movement parameters,sperm progressive motility,membrane integrity,acrosome integrity,DAN integrity,mitochondrial membrane potential and reactive oxygen species?ROS?level,and were evaluated by flow cytometry and computer-assisted sperm analysis system?CASAS?.Results,After thawing,semen freezed in the semen extender supplemented 400 ug/m L Coenzyme Q100 showed significantly higher sperm progressive motility,DAN integrity,mitochondrial membrane potential and ROS level?58.3%,96.2%,3.4 and 1414,respectively?.When catalase concentration is 200 U/mL sperm membrane integrity and acrosome integrity?44.1%and71.17%,respectively?all significantly higher than the control group(47.8%,36.3%,53.1%,93.5%and1.2,respectively.?P<0.05?.
