Systemic studies of plant regeneration of three varieties of Lolium Perenne L.(Delaware, Juventus, Taya) have been conducted. With mature embryo and matureseeds as explants, the effect of plant hormones and other nutritional substances oncallus induction and differentiation were experimented. Browning, albino, et al. werealso discussed in the research. The results go as follows.I: Mature embryo shows higher callus ratio, callus differentiation frequency, larger fresh weight, lower browning ratio and better callus quality than mature seed, while hypocotyl and radicel turn out to be the worst explants for callus induction .II.Based on the comparisons in callus ratio, callus fresh weight, browning ratio, callus quality and callus differentiation frequency, Delaware is the best of three varieties, Taya comes second and Juventus the worst.III.2~4mg/L 2,4-D is propitious to callus induction from mature embryo, and 7mg/L is favorable for mature seed. 0.025mg/L 6-BA is favorable to callus induction from mature embryo and 0 for mature seed. For callus from mature embryo, the highest induction ratios of three varieties are 98%(Delaware), 68%(Juventus) and 72%(Taya), while for callus from mature seed, they are 75%(Delaware), 40%(Juventus) and 70%(Taya).IV.6-BA has vital effect on callus differentiation.0 mg/L is propitious to the differentiation of callus from mature embryo, and the highest differentiation frequency is 90.9% for Delaware and 69.2% for Juventus.0.1~0.3mg/L is suitable for the differentiation of callus from mature seed, and the highest differentiation frequency is 87.0%(Delaware),43.3%(Juventus)and 95.0% (Taya). NAA, ZT, Cu2+can promote callus differentiation to some degree.V.Callus induction and differentiation media of each variety(mature seed as the explant) have been optimized. For Delaware, 7mg/L 2,4-D,0.1mg/L 6-BAP, 0.3mg/L NAA and 0.1g/L CaH is suitable for callus induction; 0, 0.1 or 0.3mg/L 6-BAP, 0.4mg/L NAA, 0.1mg/L ZT and 8.96mg/L Cu2+ is suitable for callusdifferentiation. For Juventus,7mg/L 2,4-D and 0mg/L 6-BAP for callus induction, and the callus differentiation media requires further study. For Taya, 7mg/L 2,4-D and 0mg/L 6-BAP for callus induction, 0.2 mg/L 6-BAP is fit for callus differentiation. The other fixed components for the media mentioned are MS basic medium added with 30g/L sucrose and 8g/L agar, and pH of 6.0. Vl.With the utilization of active carbon and Lactalbumin Hydrolysate Power, the callus differentiation frequency can be increased about 2% and the browning ratio can be decreased about 10%~ 18%.
