Leptin receptor is a member of the class I cytokine receptor family, it plays an important role in regulating body energy stores and energy metabolism. Since OBR plays an important role in leptin functioning signal transduction and it may has direct effects on the deposition of adipose tissues and the body weight, it can be considered as a candidate gene in the current study of chicken fatness deposition. The present study was designed to detect the expression rules of chicken OBR gene to discuss whether the difference of gene expression is associated with fat deposition in various gallus breeds or not. The patio-temporal expression of OBR gene of Broilers was detected by Northern-blot and RT-PCR in different growth phases. Broiler and Layer diets were fed to AA broilers and Hyline layers in a feeding experiment. Sampling liver tissue in different growth phases (new hatch, 2, 4, 6, 8 and 10 wk of age), the expression level of chicken OBR gene was detected by semi-quantitative RT-PCR in liver. The expression level of hypothalamus, intestine and adipose tissues of various gallus breeds were detected by semi-quantitative RT-PCR in different growth phases. The main results are as follows:⑴ Results of RT-PCR and Northern-blot analysis showed that chicken OBR gene was expressed in brain, leg muscle, breast muscle, gland stomach, intestine, adipose, liver, heart and hypothalamus in Broilers of 2, 6, and 12 wk of age. The OBR gene expression level was different in various tissues, and it was higher in hypothalamus, brain and heart and lower in breast muscle and leg muscle.⑵ Detecting expression level of chicken OBR gene in liver tissues of AA broilers and Hyline layers in different growth phases, results indicated that expression level of chicken OBR gene in various growth phases are different between Broilers and Layers. The expression level of OBR gene was higher in Broilers than that in Layers at hatch, 2, 6, 8 and 10 wk of age birds, but the results of 4 wk of age was opposite.⑶ Detecting expression level of chicken OBR gene in liver tissues of AA broilers and Hyline layers fed with broiller's and layer's diets, results indicated that expression level can be affected by energy level in diets, the higher energy in diet will enhance the expression level of OBR gene in Broilers and Layers.⑷ In various growth phases (8, 10, 12 wk of age ), expression level of chicken OBR gene in hypothalamus was different between Broiler and Baier breeds. Expression level of chicken OBR gene was higher in Broilers than that in Baiers at 8 and 12 wk of age, but the results of 10 wk of age was opposite.⑸ In various growth phases (2, 6, 12 wk of age), expression level of chicken OBR gene in intestine and adipose tissues was different between Broiler and Baier breeds.
