Studies On Micropropagation Techniques Of Pistacia Vera L.and Korla's Pear

Through the studies on regeneration of in vitro culture of Pistacia vera L.and species of Korla`s pear sha-01, the proper explant induced directly and the time of induction were chosen, and the main influencing factor of multiplication, rooting ,training were evaluated .At the same time ,some improving methods were put forward . Technical system of tissue culture and rapid propagation of Pistacia vera L. and sha-01 was established .The main achievements were as follows: The survival rate of shoot explant of pistacia vera L. was higher than seed of witch .MS+BA2. 0(mg/L ,follows as the same )+NAA0. 1+3.0%Sugar +0.7% agar was most suitable to initiatory culture of the explant. The survival of shoot explant of sha-01 was higher than that in summer and autumn,B5+BA 0. 5+GA 31.0+3.0%sugar +0.7%agar was most suitable to initiatory culture of the explant .MS medium was most suitable to pistacia vera L., with BA2. 0+IBA0. 05 was suitable to subculture of adventitious bud, the time of subculture was 30d .B5 medium was most suitable to sha-01. with BA0. 5+IBA0. 1 was suitable to subculture of adventitious bud , the time of subculture was 40d. Technical system of rooting of the adventitious shoots was established. 1/2MS+IBA2.0 for rooting of pistacia vera L. For sha-01, two-step-rooting method is better than one-step-rooting method. That is, Firstly put adventitious shoots into 10mg/L IBA solution for 24 hours , and then transferring the shoots into 1/2B5 medium without any hormone but with 0.3% AC to rooting . Adding 0.3% AC into the medium promoted on sha-01's rooting, but not for pistacia vera L.The environmental conditions were main factors of training and transplanting of the rooting shoots. The rooting shoots must be trained for 10d in the light with the sealing material before transplanting. And then transferred into vermiculite:perlite 1:1 medium with 85%-100% humidity, then the rooting shoots were transferred into the pots after 30d.
