Studies On Primary Culture Of Pistacia Vera L.and P. Chinensis Bge.

Pistacia vera L . , one of the nuts that has the high economic and nutritional value , has excellent characteristics of enduring drought and barren. At the present time, it mostly propagated by graft of wattle. In foretime, Pistacia vera L . had been neglecting by people and mainly relied by importation. In recent years, Xinjiang, GanSu, Beijing, YunNan, ShangDong, HeNan etc. have succeeded in introduction, but production is restricted by limited propagating materials. The purpurse of this paper is to use stem segments, seeds etc. to research the problems, such as the choice of feasible explants, browning , contamination , and germination in the primary culture of Pistacia vera L . and its rootstalk -Pistacia chinensis Bge. .The results are as follows:1. The explents of Pistacia vera L.1) The most suitable time for picking the explant should be April and May,also the explants can be collected at September or October;2) The middle and the nether parts of the stem are more suitable for tissue culture;3) The stems should be sanitized in 0.1% Mercuric Chloride for about 10 minutes;4) Comparing with several factors that affecting the browning reaction , we have obtained the suitable medium:l/2DKW+6-BA0.5mg/l+IBA0.5mg/l+GA33.0mg/l+PVP1.5g/l+Folicacid+lmg/l+ LH300mg/I+CalciumD-pantothenatelmg/l+Agar6.5g/+D-Glucosel5g/l, PH: 5.8-6.2 .5) There is no relative comparison between the activities of polyphenol oxidase in the stems from the seven years' parts and from the one years' ,and there is no positive or negative relationship between the browning condition and the activities of polyphenol oxidase.6) There are three cultivation models can be used: dark cultivation under 20℃ , dark cultivation under 25℃,and faint light cultivation under 20℃.7) The sanitization time should be 10min;8) The seedlings should be sterilized in 0.1% Mercuric Chloride for about 7min;9) The first culture medium of the seedlings should be:6-BA0.5mg/l+ IBA0.05mg/l+ NAA0.05mg/l+ GA31mg/l and 6-BA4.0mg/l+ IBA0.02mg/l+ NAA0.02mg/l.2. The explents of Pistacia1) The best time for picking the stems should be May and June;2) The sanitization time should be between the time of 10 to 12min;3) The basic medium should be l/2DKW,and the concentration of PVP should be 1.3 g/l;4) The best ratio of hormones should be 6 —BAlmg/L+IBA0.1mg/L+NAA0.1mg/L;5) The activities of polyphenol oxidase in the stems from the seven years' parts picked from Pistacia are higher than from the one years' ,but there is no positive ornegative relationship between the browning condition and the activities of polyphenol oxidase.6) The seeds of Pistacis should be treated in concentrated sulphuric acid for about 24h ,and should be cultivated in the dark;7) The sanitization time should be 15min;8) Higher concentrated cytokinin is benefited for obtaining the strong seedlings ,and the medium should be:l/2DKW+6-BA4mg/l+IBA0.05mg/l+NAA 0.02mg/l+D-Glucosel 5mg/l+Agar 6.5mg/l + Folic acidlmg/1 + Calcium D — pantothenatelmg/1 + PVP1.3mg/l and l/2DKW+6-BA4mg/l+IBA0.1mg/l+NAA 0.05mg/l+ D-Glucose 15mg/l+ Agar 6.5mg/l +Folicacid lmg/H-CalciumD—pantothenate lmg/l+PVP1.3mg/l.
