The protocol of high efficiency plant regeneration was intended for fine plant genetic transformation. In this exprement ,in vitro culture of bulb and filament of Lilium regale as explants and plant regeneration in Lilium regale were investigated. The results were shown as follows: l.Fine green callus using bulb as explant with differentiation rate of 93.33% was formed on the 1/2MS midium sumplemented with BA2.0 mg L-1+NAA1.0 mg L-1. More adventitious bud was also found in this medium. On the 1/2MS midium sumplemented with BA2.0 mg L-1+NAA1.0 mg L-1, adventitious bud also was found and subculrured many generations 2.During subculture , clusters of bud can be found on the 1/2MS midium sumplemented with BA1.0mg L-1+NAA0.1 mg L-1.3.After getting plantlets,the tender plantlets were rooted effectively on the 1/2MS midium sumplemented withNAA0.01mg L-1+IBA0.1mg L-1with rooting rate of 85.2%,average number of root was 5.5. 1 / 2MS was thought right as rooting medium. NAA0.1mg L-1and IBA0.1mg L-1 were thought as better hormone combination.4..Adding BA induced directively adventitious bud,while adding 2,4-D restrained it. 5. During field practice,through strong illumination and culture in open bottle,the survival rate could be improved.Substrate holding perlite and vermiclite with scale by 1 to 1 was optimal.
