Study On Technique Tissue Culture In Several Rhododendron

In this study, the seeds of R . protisum var giganteum Forrest et Tagg, , R . anne Franch and R . megacalyx Balf.f.et Ward germinated under aseptic conditions ,then the shoots and leaves from Rhododendron's tube seedlings were used as explants; the culture in vitro of stem and shoot tip from R.. argyrophyllum Franch had been reported,but the culture of its leaf had not,so the leaves were chosed as explants. By means of the orthogonal experiment design, analysis of variance and multiple comparisons , the best media for serval species' different explants in vitro culure were chosed .The factors influencing Rhododendron regeneration including genotypes ,basic media , TDZ, AC were analayzed, the results were as follows:(1) In the germination trial, Read with 1 .00mg/L GAs not only shortened the germination time of R . protisum var giganteum Forrest et Tagg and R . anne Franch but also enhanced the germination percentage and promoted seedling growth. The higher germination percentage and better seedlings of R . megacalyx Balf.f.et Ward could be attained only in Read, whereas GAs played an opposite role.(2) 6-BA did harm to R . protisum var giganteum Forrest et Tagg. TDZ had more effective action on the multiplication of R . anne Franch and the callus induction of R . megacalyx Balf.f.et Ward than 6-BA, KT.(3) The best sub-culture medium for in vitro R . protisum var giganteum Forrest et Tagg was WPM medium containing 0.40mg/L TDZ and 0.05mg/L IBA with multiplication rate of 3.25, shoot induction rate of 100%, shoot differentiation rate per microcutting of 2.25.The best sub-culture medium for in vitro R . anne Franch was Read medium containing 0.30mg/L TDZ and 0.05mg/L NAA with multiplication rate of 4.20, shoot induction rate of 100%, shoot differentiation rate per microcutting of 3.20.(4) For R . protisum var giganteum Forrest et lagg ,best result in callus induction wasobtained on WPM supplemented with 1.50mg/L TDZ and 0.10mg/L NAA with regeneration rate of 100%, 190.23mg per piece; for/? . anne Franch, best result in callus induction was obtained on WPM supplemented with 1 .OOmg/L TDZ and 0.20mg/L NAA with regeneration rate of 100%, 65.28mg per piece; for R. argyrophyllum Franch, best result in callus induction was obtained on Read or WPM supplemented with l.Omg/L TDZ and 0.20~0.40mg/L IBA , number 30 with regeneration rate of 67,50%, 39.35mg per piece.(5) The best medium for adventitious shoot induction from leaf explant of R . anne Franch was WPM containing 1.50mg/L TDZ and 0.20mg/L IBA with regeneration rate of 77.50% and 2.50 adventitious shoots per leaf explant;for R. argyrophyllum Franch, the best medium was Read containing l.OOmg/L TDZ and 0.20mg/L IBA with regeneration rate of 75.00% and 4.00 adventitious shoots per leaf explant.(6) The callus of R . anne Franch multiplied but did not redifferentiate, but that of R . protisum var giganteum Forrest et Tagg , R . megacalyx Balf.f.et Ward and R.. argyrophyllum Franch did. The best media for them were respectively as follows: The best medium for the callus multiplication of R . protisum var giganteum Forrest et Tagg was: WPM+TDZ1 .00mg/L+NAA0.05mg/L with multiplication rate of 1.67. For R. anne Franch, the best medium was WPM+TDZ2.00mg/L4-NAA0.10mg/L with multiplication rate of 2.30.For R . argyrophyllum Franch, the best medium was : Read+TDZl .50mg/L + IBA0.10mg/L with multiplication rate of 3.50. The best media for the callus ^differentiation of R..argyrophyllum Franch and R . megacalyx Balf.f.et Ward were respectively Read+ TDZ1.50mg/L+IBA0.10mg/L with redifferentiation rate of 40.00%, 5.00 adventitious shoots per piece and Read+TDZ0.30 mg/L+NAA0.01mg/L+AC0.20%,in my trial the best result was number 75 with redifferentiation rate of 90.00%, 22.50 adventitious shoots per piece.(7) Adventitious shoots of Rhododendrons formed on TDZ-contained medium were short ,so it was essential for them to be strengthed .
