The Study Of Tissue Culture And Rapid Propagation Of Ananas Comosus Sp.

Ananas comosus sp. is one of the improved varieties of pineapple. In view of factorize, a rapid propagation method of tissue culture in Ananas comosus sp. was established, which will resolve the problem of shortage in seedling of Ananas comosus sp. Using the root, stem and leaves as explants to induce the tufted sprouts, Callus, adventitious buds, and roots was studied in this paper. The results were as follows. The optimal explants is stem with axillary or tip buds, gathered in In March and April or Sepstember and October. Explants were sterilize in 0.1%Hgcl2 for 10 minutes after washed in tap water for 1~2 hours and sterilization of 70%alcohol for3~5secends, then inoculated it on tufted sprouts-inducing culture medium. During the induction, supplementing 0.1% activated carbon in the medrun culturing in the dark or transfering new medium could prevent browning and improve the regeneration frequency. Adventitious bud inducing culture medium was the MS medium with 2.0mg/L BA,0.1mg/L NAA and 3% sugar. The optimal callus inducing culture medium was the MS medium with 2.0mg/L NAA,0.5mg/L KT and 3% sugar. The roots, stems and leaves can be induced callus. But the stem was the optimum explants. If decreased the plant growth regulators step by step during the subculture, the shoots could grow stronger and stronger. Further more, the variability will decline. Then, the shoots were transferred to the 1/2 MS medium contained 0.1mg/L NAA, 0.1mg/L IBA and 2% sugar to induce roots. Plantlets transplanted on media contained peat: coconut husk: ruby mica: loess =4:1:1:2. Finally, the survival rate could reach above 90%. Theoretically, the coefficient of propagation of a shoot tip could reach 5 for 20~30days, so the proliferation times of a shoot tip could reach 3×108 for a year. It was a effective method in numerous proliferatin of pineapple.
