A simple plant regeneration system of Euonymus fortunei 'Orbiculatus' was established in this experiment after succeeding in inducing callus regenerating plantlets rooting and transplanting experiment.In this paper, the stem and leaves of Euonymus fortunei 'Orbiculatus' were cultivated in the Murashige and Skooge's medium supplemented with different kinds of hormones and nutriment. The effect of 2,4-D, 6-BA, NAA, IBA and culture conditions for tissue culture and plant regeneration of Euonymus fortunei 'Orbiculatus' was described. The results indicate as follow:The frequency of callus induction was different when stems or leaves were cultivated in MS medium supplemented with combinations of hormones. The optimum callus inducing medium for stems was MS+6-BA1.0~2.0mg/L+2,4-D0.05 mg/L, for leaves was MS+6-BA1.0~1.5mg/L+2,4-D0.1mg/L.On the experiment of subculture, the best medium for stems was 6-BA1.5mg/L+IBA0.05~0.1mg/L or 6-BA1.5 mg/L+NAA0.05mg/L; the best one for leaves was 6-BA1.0-1.5 mg/L+ IBA0.05~0.1mg/L or 6-BA1.0-1.5 mg/L + NAA 0.05-0.lmg/L. It showed that the medium supplemented with sugar 2.0% was best for the growth of leaves callus. On the experiment of plant regenerating, the best medium for plant regenerating of stems was MS + 6-BA1.5mg/L+NAA0.05mg/L or MS + 6-BA1.3mg/L+IBA0.1mg/L. It showed that the medium placed in light supplemented with sugar 3.0% was best for the growth of callus. On the experiment of Euonymus fortunei subculture, the best medium for shoot multiplication and growth was MS + 6-BA1.0mg/L + NAA0.01lmg/L. The best medium for rooting was l/2MS+NAA0.5mg/L.
