A series of methods and techniques were studied about shoot tip culture, bulb forming and dormancy breaking in vitro, scale cutting and commercial bulb production in Oriental Hybrid Lilium ( 'Sorboune' , 'Acapulco' and 'Siberia' ).1 .Meristem culture of Oriental Hybrid Lily through its scales was studied in MS medium with different external hormone treatments. Results are as follows: The best medium for bulblet induction through scales is MS+6-BA1.0mg/L+NAA 0.2mg/L; The best medium for shoot tip induction and differentiation is MS+6-BA1.0mg/L+NAA 0.5mg/L.2.Bulb growth from adventitious buds in vitro was observed in MS medium under different sucrose concentrations and light intensities. The results show that the optimum sucrose concentration for bulblet growth in vitro is 75g/L and darkness favours the bulblet.3.The experiments of dormancy breaking and germplasm conservation were performed in four temperature conditions(10℃, 5℃, -1℃, -5℃). The results indicate that: Low temperature is very important for the growth of tissue culture plantlet, the best low temperature condition is 5℃ for 8 weeks and the better is 10℃ for 12 weeks. The best temperature for germplasm conservation is -1℃ and cryopreservation is not suitable for lily bulbs.4. We analyzed the effects of different humidity, media, cultivaxs and different parts of scale on germination, the effects of different seeding time on bulb growth in different cultivars and the effects of different cultivation pattern on commercial bulb growth. A series of methods and techniques were established from scale cutting, bulblet propagation to commercial bulb production. Our research shows that :The germination is best when outside scales are under 25℃, 80% RH and 2:1 of peat to perlite, and there is no obvious difference between cultivars. The bulblets' diameter could reach 6~8cmwhen the buds were planted in seedbed at early March or early September. Autumn is beneficial to the growth of bulblets and there are no differences between cultivars. Part of bulbs could reach the standard of commercial bulb and could be applied in production if the cold treated buldlets were planted at early March and harvested in November or postponed to next year's July.
