The Research On Extraction And Medicinal Effects And Pharmacokinetics Of The Total Effective Constituents Of One Chinese Herbal Medicine

The research is on the extract and medicinal effects and pharmacokinetics of one compound Chinese herbal medicine which can control and prevent aquatic animals from Bacterial Septicamia disease. An orthogonal experiment (L934) was made to check the optimal extraction of this compound Chinese herbal medicine. The medicinal effects of the total effective constituents extraction on controlling and preventing Carassius auratus from Bacterial Septicamia disease was studied. The pharmacokinetics of the total effective constituents extraction in Carassius auratus was studied by drug cumulative method.Experiment One: An orthogonal experiment (L934) was made to check the optimal extraction Condition in the indexs of the content of flavones , anthrapuinone and gallic acid, those are the effective constituents of the Chinese herbal medicine. The four factors of the orthogonal experiment (L934) were alcohol concentrate, times of farinose herbal medicine, extraction times and time for one extraction. The bacterial inhibition effects of total effective constituents extraction were studied. The results show that 65% ethanol, 12 times of farinose herbal medicine , 76℃ , 3 times extract and 3h for one extract was the optimal condition. Extraction times was the biggest factor of the four factors. The content of total flavones, total anthrapuinone and gallic acid in the total effective constituents extraction were 14.768,1.746 and 17.432mg/g. The bacterial inhibition effect of the oral total effective constituents extraction was very good and the diameter of bacterial inhibition loop was 3.6cm, and the bacterial inhibition effects were also good for the 5 times and 10times diffuse of the oral total effective constituents extraction and the diameters of bacterial inhibition loop were 2.4cm and 1.4cm.Experiment Two: The total effective constituents extraction was addicted into feeds and different additive of 0 0.5% 2% drug baits were made. The medicinal effects on controlling and preventing Carassius auratus from Bacterial Septicamia disease was studied. In the different course of disease the serum physiologic and biochemical indexs were detected. The acute toxicity of the compound Chinese herbal medicine was also studied. The results show that the serum physiologic and biochemical index of the Carassius auratus were unmoral after the fish was artificial infection by Bacterial Septicamia disease bacteria and the serum glutamic-oxalacetic transminase , serum creatinine.serum uric acid , serum urea nitrogen , serum granulocyte cell ,serum creatine phosphokinase ,white blood cell were on the rise ,serum cholesterol, serum total protein , serum albumin , serum globular proteins , red blood cell ,serum hemoglobin were decline.
