The Study On The Prevention And Treatment Of Compound Chinese Herbs "DUJUNSHA" To Infectious Bursal Disease (IBD) In Chicken

Infectious Bursal Disease is an acute and contactile contagion caused by IBDV, which harm mainly the chichens, and bring enomous losses to poultry husbandry in all world. Chinese herbal medicines have many functions, such as inhibiting bacterium, resisting virus, eliminating inflammation, procreating interfeorn,regulating the body immune function ,increasing the body resistility and play an important part in prevention and treatment virus .This objective in reseach is to observe the effect of compound Chinese herbs "DUJUNSHA" in the prevention and treatment of Infectious Bursal Disease (IBD) in chicken. The study consisted of three parts.The prescription of compound Chinese herbs "DUJUNSHA" was designed based on proved recipes and Chinese medicine theories. The method of water extraction was used to make compound Chinese herbs "DUJUNSHA". The safety experiments were performed in rabbits and mice , including the test of heat source and bacteria, allergy test and hemolysis test. The results showed that compound Chinese herbs "DUJUNSHA" accorded with all the quality criterions of Chinese herbal medicine regulated by " The pharmacopeia of the People's Republic of China" . So it could be used in clinical tests.The distilled Chinese herbal medicine liquid was diluted 1g/ml, 0.5 g/ml, 0.25 g/ml respectively. The powerful IBDV ,GX-8/99, was diluted to 100 EID₅₀ .Then every kind concentration of Chinese herbal medicine liquid was mixed with IBDV diluted with equimutiple .Then they were placed in 37℃ water for 2 hours .The 0.2ml virus-Chinese herbal medicine liquid mixture was inoculated in allantoic antrum of 9-10 days chikens,accroding physiologycial salt water and IBDV to masculine and minus teams repectively .The results showed :the 83.3% of survival rate of chicken embryo was highest,when the concentration of Mixture â…¡ was 1 g/ml ,however ,the 33.3% of survival rate of chicken embryo was lowest when the concentration of Mixture â…  was 0.25g/ml .The survival rate of chicken embryo was above50% in other groups. All chicken embryo of virus team were dead after 120 hours of inoculation. The results showed: three kinds of mixtures have good functions on resisting virus and prolonging the life of chicken embryo and the effect resisting virus of Mixture II was better than that of Mixture I and III .The bacteriostasis in vitro on E.coli gallinacean and S.pullorum of every Chinese herbal medicine and compound Chinese herbs "DUJUNSHA"were observed by flat meliorated way. The results showed: every herb has obvious effect in inhibitting E.coli gallinacean and S.pullorum.the minimum anti-bacteria concentration was from 0.25g/ml to 0.5g/ml. The minimum antibacteria concentration of mixture was lower than that of individual herb, and especially the effect of Mixture Two was better. To E.coli gallinacean and S.pullorum the minimum antibacteria concentration was 0.25g/ml and 0.125g/ml respectively.Through the observion on the test I and II ,the effect of Mixture II on bacteriostasis and antivirus was better than that of Mixture I and III .So the Mixture II was introduced to carry through the antivirus experiment in chikens.Healthy chickens (n=320) were randomly divided into 4 groups: prevention group(Mixture II), treatment group (Mixture II),positive and negtive control group. The chickens of prevention group were fed Chinese herbal medicine "DUJUNSHA" in 3 days before inoculation of virus. The chickens of treatment group were fed Chinese herbal medicine "DUJUNSHA" after they were inoculated IBDV. The chickens of positive control group were only inoculated IBDV. The chickens of negtive control group were neither inoculated IBDV nor fed Chinese herbal medicine "DUJUNSHA". The clinical symptom and death were observed every day .The index of immune organs (thymus, bursa, spleen) was mensurated in 1 day before inoculation of IBDV and 1,7,14,2l,28,35days after inoculation of IBDV. The WBC quantity and the rate of ANAE masculine lymphocyte were determined in lday before inoculated IBDV and l,3,7,15days after inoculated IBDV. The results showed: Chinese herbal medicine "DUJUNSHA" can availably prevent IBDV from hurting immune organ (thymus, bursa, spleen) and maintain the opposite stability of ANAE positive lymphocyte .So the body immune function and the ability of resistivity and prevention on IBDV was improved obviously.
