In paper, the seeds of Triticum aestivum L.(Jing 411) were treated by different concentrations of SAM, the methyl-donor, to understand the effect of DNA methylation on the growth and development of the primary root under the levels of morphology, cytology and molecular biology.The results are as follows:(1) The growth of wheat primary root was inhibited by the treatment of SAM, and the inhibition was dependent on the dose of SAM as well as the treated time.(2) The increase of DNA methylation was correlated positively with the concentration of SAM and the time of culture, too.(3) There was negative correlation between the mitotic index and the extend of DNA methylation. That is to say, the mitotic index decreased while the the extend of DNA methylation increased.(4) 500 spots on the gels with a pH3-10 gradient and 600 spots on the gels with a pH4-7 gradient were observed, respectively, when the gels were stained by Coomassie blue and analyzed by ImageMasterTM2D Platinum software (version 5.0). Comparing the control with the treatment in 20μmol/L and 80μmol/L SAM, respectively, about 17 and 13 proteins were checked up-regulated, and 22 and 29 spots were down-regulated. There were no differentially expressed dots depicted. The sequences and functional information of these proteins needed more investigation.(5) There was no definite result of Northern blotting because of the selection of the probe. But we can inferred by the results from morphology and cytology analysis that the translation and expression of the genes that control the cell division and elongation were likely to be inhibited, and then the mitotic of the wheat root-tip meristematic cell and the growth and development of its primary root were influencd.
