On the basis of AU-PAGE method, SRAP and SSR technology, 20 watermelon hybrids and 2 watermelon hybrid combinations and their parents were studied on genetic diversity and purity identification. The related technology systems were modified and improved. SRAP method on purity identification of watermelon hybrids was established. The results were as follows: (1) SRAP could be applied on watermelon hybrids purity identification:3 primer pairs(ME-6/EM-2, ME-6/EM-3, ME-2/EM-1) showed polymorphic on XiFeng1 and its parents. 3 primer pairs (ME-1/EM-2, ME-1/EM-3, ME-8/EM-6) showed polymorphic on XiFeng2 and its parents. 20 watermelon hybrids could be identificated by 19 polymorphic primer pairs. Purity identification of 4 watermelon hybrids with plant testing method and SRAP (primer pair: ME-2/EM-5) technology showed the purity accordant rate between two methods ranged from 93.8~96.8%, SRAP result was a lttile lower than plant testing result. It showed SRAP had more ablity to identify purity than plant testing method. (2) SRAP technology system fitted for watermelon seed DNA was established: regular DNA extraction methods was fitted for SRAP identification, changed leaf DNA extraction into seed DNA extraction, modified SRAP reaction system(25ul reaction system contained DNA template 30ng, primer pairs 5pmol, Mg2+ 3mM,TaqE 0.5U, dNTPs 0.2mM) and testing technology of reaction products.
