Rice is the largest crop in our country. Its output and quality are influenced by the quality of the seed and the purity of the seed is the most troublesome problem in the control of seed quality. SDS-PAGE technique was applied to compare and analyze five hybrid rice and their three lines and nine different F1 hybrids, which are widely cultivated in Fujian province at present, A simple, rapid and economical method was selected for rice purity identification and a standard procedure was established. At the same time, the purity of several hybrids was also analyzed, discussing its practicability and prospect. Major results were as follows:1. The salt-soluble of three different hybrid rice and their three lines were analyzed based on SDS-PAGE. The Results showed that the polymorphism of salt-soluble proteins was high. The Bands of three lines and its F1 were almost the same but there were obvious differences in number, width and color. Based on its high resolving power and stable specific bands, it could be used for purity identification. At the same time, eight different varieties and the same variety Tyou 551 and Jinliangyou36 planted in different planting areas were analyzed too. The experiment showed that .the quantity of the bands, position distribution and depth of different hybrid had obvious differences. Thus, it showed the differences of hybrid rice seeds of different varieties. The seeds planted in different areas or years had the same bands, not influenced by the land fertility level. The same sample Jinliangyou36 was divided into two. The samples' purity planted in Hainan was 97. 6% and 95. 2%. The purity of 100 seeds collected at random for electrophoresis detection were 99% and 95%. There were identical results between them, indicating the SDS-PAGE technique used for rice purity identification was reliable and accurate. Experiments also demonstrated that SDS-PAGE is a method which is simple, rapid, effective and inexpensive. This method can be applied in the identification of authenticity and purity of the seed.2. The salt-soluble proteins of germinating seeds were analyzed on four different hybrid rice and their three lines by SDS-PAGE. The experiment showed that: the salt-soluble proteins of germinating seeds had high polymorphism and can be applied in the identification of authenticity and purity of the seed. But, compared with common seeds, germinating seeds were not the suitable materials for identification of authenticity and purity of the seed.
